高密度培养大肠杆菌酶法合成L-天冬氨酸

摘要 运用正交实验对大肠杆菌HY-05C的培养基进行优化,获得最佳培养基(g/L):富马酸铵10,玉米干粉8,酵母粉2,蛋白胨7,氯化钠5,磷酸二氢钾1,硫酸镁0.2,通过单因素实验确定培养基的最适p H 6.5,最佳接种量为1‰(V/V)。一、二级种子培养时间为6-8小时,三级培养液8小时菌体量和酶活最高。菌体量达到2.49×108cfu/m L,提高了40~50%;酶活达到7500U/m L,提高了70-80%。 Orthogonal experiment has been employed to optimize the medium for L-aspartic acid production by Escherichia coli HY-05 C.The optimal fermentation medium consisted of the following( g/L):ammonium fumarate 10, corn steep liquor powder 8, yeast powder 2,peptone 7,Na Cl 5,KH2PO4 1,Mg SO4 0.2,The optimalest p H is 6.5 according to the amono-factor experiment,The optimalest inoculation amounts are 1‰(V/V).The incubation time of fi rst and second order seeds 6-8,The three period culture medium has the highest biomass and enzyme activity at eight hour.the biomass that improved by 40-50% reached to 2.49×108cfu/m L;Enzyme activity that improved by 70-80% reached to 7500U/m L.
