木丹颗粒对糖尿病大鼠背根神经节蛋白表达及血清p38MAPK的影响 被引量:1

The Influence of Mudan Granule on Protein Expression of Dorsal Root Neural Ganglia and Serum p38MAPK of Rats with Diabetes

摘要 目的:观察木丹颗粒对链脲佐菌素(STZ)诱导的糖尿病大鼠背根神经节中磷酸化的p38MAPK(p-p38MAPK)和Cyt C的蛋白表达及血清中p38MAPK的影响。方法:将8只正常的大鼠分入正常对照组,将造模成功的36只大鼠随机平均的分入模型组、木丹颗粒组、甲钴胺片组,每组13只。采用STZ诱导复制糖尿病模型,正常对照组和模型组灌胃生理盐水,甲钴胺片组和木丹颗粒组分别灌胃甲钴胺片和木丹颗粒,对比各组大鼠血清中的p38MAPK水平,对比各组大鼠背根神经节中p-p38MAPK和Cyt C的蛋白表达情况。结果:模型组大鼠血清中的p38MAPK水平显著性的高于正常对照组,背根神经节中p-p38MAPK灰度值和Cyt C灰度值显著性的低于正常对照组(P<0.05);木丹颗粒组大鼠血清中的p38MAPK水平显著性的低于模型组,背根神经节中p-p38MAPK灰度值和Cyt C灰度值显著性的高于模型组(P<0.05)。结论:木丹颗粒保护糖尿病大鼠背根神经节的作用机制可能为抑制线粒体中Cyt Cd蛋白释放、降低血清中p38MAPK水平、抑制背根神经节中p38MAPK磷酸化。 Objective: To observe the influence of Mudan Granule on phosphorylated p38 MAPK and protein expression of Cyt C of dorsal root neural ganglia and serum p38 MAPK of rats with diabetes induced by STZ. Methods: Eight normal rats were chosen into the normal control group. After successful modeling,36 rats were randomly divided into the model group,the MDG group,the mecobalamin tablet group,with 13 cases in each group. First,the diabetes model was copied by using STZ inducement; The rats in the normal control group and the model group were given normal saline through administration by gavage; The ones in the mecobalamin tablet group and the MDG group were given mecobalamin tablet and MDG respectively through administration by gavage.Then each group's serum p38 MAPK level,phosphorylated p38 MAPK and protein expression of Cyt C of dorsal root neural ganglia were compared. Results: In terms of serum p38 MAPK level,the model group was much higher than the normal control group. In terms of gray values of p-p38 MAPK of dorsal root neural ganglia and Cyt C,the model group was much lower than the normal control group( P〈0. 05). In terms of serum p38 MAPK level,the MDG group was much lower than the model group. In terms of gray values of p-p38 MAPK of dorsal root neural ganglia and Cyt C,the MDG group was much higher than the model group( P〈0. 05).Conclusion: MDG can protect dorsal root neural ganglia of rats with diabetes,and its working mechanism is to suppress Cyt Cd protein release of mitochondrial,reduce serum p38 MAPK level,and suppress p38 MAPK phosphorylation of dorsal root neural ganglia.
