盐含量和高压灭菌对培养基酸碱度和凝固状态的影响 被引量:3

摘要 将MS,B5和W培养基配制成有机元素含量不变,无机盐含量分别为100%,75%,50%,25%,0的培养基,分别加入不同体积1 mol·L-1的Na OH溶液,测定高压灭菌前后培养基p H值,记录培养基凝固状态,研究不同盐含量MS,B5和W培养基高压灭菌后p H值与Na OH溶液添加量之间的定量关系,以及盐含量和酸碱度变化对培养基凝固状态的影响。结果表明,高压灭菌后,不同盐含量的培养基p H值与Na OH溶液添加量呈对数关系,培养基p H值降低幅度随盐含量增加而减小,调节培养基p H值所需Na OH溶液添加量随盐含量增加而增加,培养基硬度随Na OH溶液添加量增加而增加。 In the present study,MS,B5 and W media with unchanged organic elements and varied inorganic salt strength of 100%,75%,50%,25% and 0,were selected as materials. With addition of different volumes of 1 mol·L- 1Na OH solution,the p H value of media before and after autoclaving were determined,and solidification state of media were examined to clarify the quantitative relationship between p H value and the added volume of Na OH solution,as well as effects of salt strength on p H value and solidification state of MS,B5 and W media after autoclaving.It was shown that there was a logarithmic quantitative relationship between the added volumes of Na OH solution and p H value of media after autoclaving. The media p H value decreased as salt strength increased,while the volume of Na OH solution needed for p H adjustment increased as salt strength increased. The hardness of media increased as the added volume of Na OH solution increased.
