摘要
分别以不同品种桂花的幼胚为外植体进行组织培养,研究了不同激素条件对离体培养的桂花幼胚存活率、出芽率、幼苗平均株高以及平均丛生芽数等的影响.实验结果表明:籽银桂幼胚在1/2 MS+0.5mg·L-1 TDZ+0.1mg·L-1 NAA培养基上培养,幼胚存活率为100%、萌芽率为50%、幼苗平均株高度为3.9cm,丛生芽数达3个;莲子丹桂幼胚存活率高达100%,其幼苗平均高度为2.2cm;"庐州黄"籽银桂平均丛生芽数多达3个.此研究为桂花的大量离体繁殖提供了新的实验体系.
Young embryos from immature seeds of different varieties of Osmanthus fragran were cultured in vitro.The effects of different hormone on the survival rate,germination rate of young embryo,height of shoots and number of multiple shoots were investigated.The results showed that the survival rate,germination rate were up to 100%,50% respectively,the mean height of shoots,the mean number of multiple shoots were 3.9cm and 3respectively when the young embryos of Osmanthus fragrans cv.Ziyingui were cultured on the 1/2 MS medium supplemented with 0.5mg·L-1thidiazuron and0.1mg·L-1naphthaleneacetic acid.The survival rate of young embryos and the mean height of shoots in Osmanthus fragrans cv.Lianzidangui were up to 100%,2.2cm respectively. The mean number of the multiple shoots in"Luzhouhuang"ziyingui was 3.This study represented a new experimental system for large in vitro propagation of O.fragrans.
