摘要
目的构建后循环缺血性眩晕(PCIV)气虚血瘀证家兔模型,观察眩晕定方对循环缺血性眩晕的影响,并初步探讨其可能的作用机制。方法从40只家兔中随机选取8只设为空白对照组(KBDZ组),其余32只采用"饥饿+限量高脂饲料+颈椎旁硬化剂注射+旋转刺激"多因素方法造模。在造模进行至第4周末,选择符合模型标准的32只随机分为模型对照组(MXDZ组)、眩晕定高剂量组(XYDG组)、眩晕定中剂量组(XYDZ组)、养血清脑颗粒组(YXQN组),每组各8只。各组分别给予相应药物灌胃干预2周,实验结束时通过透射电镜观测前庭神经核组织超微病理的改变,采用诱发电位仪观测脑干听觉诱发电位(BAEP)的改变。结果电镜下观察前庭神经核组织病变严重程度依次为:MXDZ组〉YXQN组≥XYDZ组〉XYDG组〉KBDZ组。药物干预后,与MXDZ组波峰潜伏期比较,XYDG组Ⅰ波、Ⅴ波差异具有统计学意义(P〈0.01),XYDZ组、XYDG组Ⅰ~Ⅴ波IPL差异有统计学意义(P〈0.05或P〈0.01);与YXQN组比较,XYDG组、XYDZ组Ⅴ波差异具有统计学意义(P〈0.05),Ⅰ~Ⅴ波IPL差异有统计学意义(P〈0.05或P〈0.01)。结论眩晕定方可恢复脑干、前庭神经功能,其作用机制可能与改善脑干听觉诱发电位有关,从而发挥对前庭神经核组织的保护作用。
Objective To observe effect of Xuanyunding decoction(XYDD)on brain stem vestibular function in rabbits with posterior circulation ischemia vertigo(PCIV)and syndrome of qi deficiency and blood stasis,and to explore the possible mechanism.Methods Methods Forty rabbits were randomly divided into four groups.Eight rabbits was in blank control group,and the others were modeled with the compound method of starvation,high lipid feedstuff,injection of sclerosing agent beside cervical vertebra and rotation.After four weeks,24 rabbits were randomly divided into three groups:Model control group,XYDD high dose group,XYDD medium dose group and Yangxue Qingnao granule(YQG)group,8 cases in each group,with two weeks intervention.The histological change of vestibular nucleus was observed by transmission electron microscope,and brainstem auditory evoked potential(BAEP)was also determined.Results The severity of histological change was followed by model control group,YQG group,XYDD medium dose group,XYDD high dose group,blank control group in order.After intervention,there was statistic significance in preclinical wave crest of raveⅠand raveⅤ between model control group and XYDD high dose group(P 〈0.01).There was statistic significance in aveⅠto V interpeak latency(IPL)between model control group and XYDD high or medium dose group(P 〈0.05 or P 〈0.01).The differences were statically significant in waveⅤ and waveⅠto V IPL between YQG group and XYDD high or medium dose group(P 〈0.05 or P 〈0.01).Conclusion XYDD may improve the function of brain stem and vestibule by protecting vestibular nucleus and brainstem auditory evoked potential in rabbits with PCIV and syndrome of qi deficiency and blood stasis.
