摘要
目的建立检测九味肾清颗粒中黄芪甲苷含量的高效液相色谱-蒸发光散射(HPLC-ELSD)法。方法以黄芪甲苷为测定指标,色谱柱为依利特Hypersil ODS 5μm,4.6mm×250.0mm,流动相为乙腈-水(3862);流速1.0mL·min^-1,漂移管温度110℃;载气流量2.59L·min^-1。结果黄芪甲苷对照品线性范围为1.758 8-17.588 0μg,回归方程为Y=1.352 8 X+4.785 9,r=0.999 7,平均回收率为101.51%,RSD为2.91%。结论HPLC-ELSD法可以准确地进行定量分析,可用于九味肾清颗粒的质量控制。
Objective To establish a high performance liquid chromatography-evaporative light scattering detection(HPLC-ELSD)method for determining the content of astragaloside in Shenqing granules.Methods The HPLC-ELSD of astragaloside was performed on the Hypersil ODS chromatography column(4.6mm×250.0mm,5μm)with acetonitrile-water(3862)at a flow rate of1.0mL·min^-1.The drift tube temperature was set at 110℃,with a carrier gas flow rate of 2.59L·min^-1.Results The linear range of astragaloside reference substance was 1.758 8-17.588 0μg.The regression equation was Y=1.352 8X+4.785 9(r=0.999 7).The average recovery rate was 101.51%,with a RSDof 2.91%.Conclusion The HPLC-ELSD method is accurate for quantitative analysis of astragaloside and can be used for controlling the quality of Shenqing granules.
