摘要
目的探讨牛蒡子苷元对人胰腺癌细胞PANC-1裸鼠移植瘤生长的作用及其相关机制。方法建立PANC-1细胞的裸鼠移植瘤模型,将荷瘤鼠随机分为对照组和实验组,实验组按体重给予10μg/g牛蒡子苷元,对照组则给予等量1640培养液。各组均采用腹腔注射给药,1次/d,持续2周。实验过程中测量肿瘤体积。免疫组织化学法检测增值细胞核抗原(PCNA)及血管内皮生长因子(VEGF)的表达。结果末次用药后实验组肿瘤体积和质量[(264.84±51.44)mm3,(133.27±21.32)mg]明显小于对照组[(695.87±127.65)mm3,(239±27.92)mg],具有统计学差异(P<0.01)。免疫组化染色结果示实验组PCNA及VEGF的表达量(28.53±4.19,0.09±0.02)均明显低于对照组(77.81±6.05,0.31±0.05),具有统计学差异(P<0.01)。结论牛蒡子苷元可抑制胰腺癌的生长增殖,其机制可能与下调VEGF及PCNA的表达有关。
Objective To investigate the anti-tumor effect of Arctigenin on PANC-1 cell xenograft on athymicmice. Methods The models of PANC- 1 cell xenograft on athymic mice were established and were randomized into two groups with intraperitoneal injection of different drugs, experiment group (Arctigenin 10 tag/g), control group (1640 medium). The drugs were injected once a day, two weeks in all. The tumor volume and tumor weight were measured. Immunohistochemistry was performed to detect the expression of proliferating cell nuclear antigen (PCNA), vascular endothelial growth factor (VEGF). Results Compared with the control group, the tumor volume and tumor weight in experiment group were significantly decreased [(264.84 ±51.44) mm3 vs (695.87 ±127.65) mm3, (133.27 ±21.32) mg vs (239 ±27.92) mg, all P〈0.01[, and the expression of PCNA and VEGF were also suppressed [(28.53 ±4.19) vs (77.81 ± 6.05), (0.09 ± 0.02) vs (0.31 ± 0.05), P〈0.01]. Conclusion Arctigenin may possesess the anti-tumor effect of panceatic cancer xenograft at least in part via downregulating the expression of PCNA and VEGF.
