摘要
从大连海域采集的海泥样品中筛选能产碱性蛋白酶的海洋细菌。用酪蛋白平板涂布法筛选得到55株具有产碱性蛋白酶能力的菌株。分离纯化后,经酪蛋白平板点种法初筛和福林酚法复筛,得到3株具有较强产碱性蛋白酶能力的菌株DL07、DL18和DL31,其酶活分别为156.5 U/m L、129.7U/m L和179.4 U/m L。实验结果表明从大连海域获得了碱性蛋白酶高产菌株,丰富了菌种资源。菌株DL07和DL31将来可作为良好的出发菌株进行改良、驯化,探索产物碱性蛋白酶优良特性,以期适用于工业应用。
To Isolate the alkaline protease-producing marine bacteria from sea mud samples of Dalian sea area, 55 bacteria which can product alkaline protease were obtained from Casein media using spread-plate method. And after the separation and purification, 3 strains called DL07, DL18 and DL31 with stronger enzyme production capacity have been got by size comparison of the hydrolysis zone and by rescreening by Folin-phenol reagent method. And their enzyme activity were 156.5 U/m L, 129.7U/m L and 179. 4U/m L. It shows that we can get alkaline protease-producing marine bacteria from Dalian sea area, and the species resources may be enriched. Strain DL07 and DL31 may be improved and domesticated as the good starting strains.
