摘要
目的 建立五指那藤中齐墩果酸的反相高效液相色谱含量测定方法.方法 色谱柱为LiChrospherC18柱(250 mm×4.6 mm,5 μm),流动相为乙腈-甲醇-水-冰乙酸-三乙胺(65∶12∶23∶0.04∶0.02),流速1.0 mL/min,检测波长210 nm,柱温30 ℃.结果 齐墩果酸进样量在0.012 36~0.247 2 μg 范围内与峰面积呈良好的线性关系,回归方程为Y =5.13×105X -6.11×102(r =0.999 9);平均加样回收率为97.47%,RSD=2.35%(n =6).结论 该方法简便、准确、可靠,可作为五指那藤中齐墩果酸的含量测定方法.
Objective To establish an RP-HPLC method for content determination of oleanolic acid in Stauntonia obovatifoliola Hayata subsp.urophylla. Methods The RP-HPLC with Li Chrospher C18 column(250 mm × 4.6 mm, 5 μm) was used, acetonitrile-methonal-water-acetic acid-triethylamine(65∶12∶23∶0.04∶0.02) was used as mobile phase, with flow rate of 1.0 m L/min, detection wavelength at 210 nm and column temperature at 30 ℃. Results Oleanolic acid showed good linearity(r =0.999 9) in the range of 0.012 36-0.247 2 μg;regressive equation was Y =5.13 × 105X-6.11 × 102;the average recovery was 97.47%;RSD was 2.35%(n =6). Conclusion The method is simple, accurate and reliable, which can be used to determine the content of oleanolic acid in Stauntonia obovatifoliola Hayata subsp.urophylla.
