摘要
目的:建立同时测定鹿蹄草中齐墩果酸和熊果酸含量的方法。方法:采用反相高效液相色谱法。色谱柱为Kromasil C18(250 mm×4.6 mm,5μm),流动相为甲醇-水-磷酸(88∶12∶0.15,V/V/V),流速为0.9 ml/min,柱温为30℃,检测波长为210 nm。结果:齐墩果酸进样量在0.462-4.620μg时,与峰面积积分值呈良好的线性关系(r=0.99998,n=6),平均加样回收率为98.6%,RSD=2.1%(n=6);熊果酸进样量在1.064-10.640μg时,与峰面积积分值呈良好线性关系(r=0.99999,n=6),平均加样回收率为98.5%,RSD=2.2%(n=6)。结论:该方法操作简单、结果准确、重复性好,适用于鹿蹄草中齐墩果酸和熊果酸含量的同时测定。
OBJECTIVE: To establish a method for the content determination of oleanolic acid and ursolic acid in Pyrolae hor- ha. METHODS: RP-HPLC method was adopted. The separation was achieved on a Kromasil C18(250 mm×4.6 mm, 5 μm) column with mobile phase consisted for methanol-water-phosphoric acid (88 : 12 : 0.15, V/V/V) at the flow rate of 0.9 ml/min. The column temperature was set at 30℃, and the detection wavelength was maintained at 210 nm. RESULTS: The linear range of oleanolic ac- id were 0.462-4.620 pg (r=0.999 98, n=6) with an average recovery of 98.6% (RSD=2.1% ,n:6); the linear range of ursolic acid were 1.064-10.640 pg(r=0.999 99,n=6) with an average recovery of 98.5% (RSD:2.2% ,n=6). CONCLUSIONS: The method is simple, accurate and repeatable, and is suitable for content determination of oleanolic acid and ursolic acid in P. callian- thr7
