柱温变化对高效液相色谱法测定黄芩苷的影响 被引量:5

Influence of temperature on parameters of HPLC by measuring Baicalin

摘要 改变高效液相色谱(HPLC)分离柱的温度,对黄芩苷含量进行测定。随着温度的升高,黄芩苷的保留时间、峰面积、有效塔板数、分离度等参数都发生了相应变化变化,45℃时,系统压力为4.2 MPa,黄芩苷保留时间为5.193min,有效塔板数和分离度均最大,拖尾因子接近1.00,黄芩苷峰较窄且对称性好,图谱最优。 The content of baicalin was determined by HPLC while the separation column temperature was changed. The results showed that the parameters of HPLC, such as retention time, peak area, effective plate number and separating degree were correspondingly changed with the increase of temperature. At 45 ℃ ,the pressure of HPLC system was 4. 2 MPa,retention time was 5. 193, the trailing factor was close to 1.00, both the plate number and separating degree were all largest,the baicalin peak was narrow and symmetrical,and the chromatogram was optimal.
