摘要
目的观察黄茯胶囊对尿酸所致大鼠痛风性关节炎模型的影响。方法将雄性SD大鼠60只随机均分为正常组、模型组、吲哚美辛组及黄茯高、中、低剂量组。采用尿酸钠溶液注入大鼠右侧踝关节腔造成急性痛风性关节炎模型,给予正常组生理盐水。观察各组大鼠用药前后一般体征、测量受试关节周径,于末次给药后24 h检测关节周围软组织中PGE2、IL-1、IL-6的含量。结果给药3 d后,黄茯胶囊组大鼠一般体征趋于正常,黄茯高、中剂量组的关节肿胀度较模型组非常显著地降低;黄茯高、中剂量组PGE2含量较模型组显著的降低;黄茯高剂量组的IL-1含量较模型组显著降低;黄茯高、中剂量组的IL-6含量较模型组显著降低。结论黄茯胶囊对痛风性关节炎有抑制关节肿胀和抗炎、镇痛的作用,其作用机制与降低关节周围软组织中炎症因子PGE2、IL-1、IL-6含量有关。
OBJECTIVE To observe the effects of Huangfu capsule on gouty arthritis in rat caused by models uric acid. METHODS The 60 male SD rats were randomly divided into normal group, model group, the indomethacin group, Huangfu capsule ( high, medium and low dose) groups. The uric acid sodium solution injected into the right ankle joint cavity caused acute gouty arthritis of rats model, and the normal group was given the physiological saline. The general physicay signs was observed and joints' s circumference was meas- ured in each group medicine before and after,in the last 24 h after dosing to detect joint in the surrounding soft tissue. And the contents of PGE2, IL - 6, IL - 1 were measured. RESULTS For three days after dosing, to the general symptoms Huangfu capsules group rats tend to be normal, Huangfu capsule high, medium dose group rats of joint swelling degree was very significantly lower, compared with the model group. Huangfu capsule high, medium dose group of PGE2 levels significantly lower in the model group. Huangfu capsule high - dose group of IL - 1 levels significantly lower in the model group. Huangfu capsule high, medium dose group of IL - 6 levels was signifi- cantly lower in the model group. CONCLUSION Huangfu capsule has inhibition on gouty arthritis joint swelling and the effect of anti - inflammation and analgesia. Its mechanism of action is releated to the reduction of joint inflammation factor PGE2, IL - 6 and IL - 1 in the surrounding soft tissue.
