摘要
为探讨KRT31基因是否是影响和田羊羊毛细度的候选基因,试验以随机采集的400只和田羊个体的血样为材料,利用PCR-SSCP、DNA测序等技术研究了和田羊KRT31基因的多态性及其与羊毛细度的关联性。结果表明:和田羊KRT31基因具有多态性,存在3种基因型:EE、EF和FF,且处于Hardy-Weinberg平衡状态(P>0.05),多态信息含量(PIC)为0.369 8,属于中度多态(0.5>PIC>0.25)。KRT31基因编码区片段在碱基序列52 bp(C→T)处发生突变。关联分析表明:和田羊不同基因型之间的羊毛细度差异均不显著(P>0.05)。
In order to explore if KRT31 gene is a candidate gene affecting the wool fineness of Hetian sheep, 400 individual blood samples were collected and analyzed for the information of its polymorphism and its correlation with the wool fineness by PCR-SS-CP analysis and DNA sequencing technology. The results showed that there was a polymorphism in KRT31 gene; the KRT31 gene had three genetypes(EE, EF, FF) and was in the Hardy - Weinberg equilibrium state (P〉0.05), the polymorphism information content (PIC) was 0.369 8, belonging to moderate polymorphism(0.25 〈 PIC 〈 0.5 ). The sequencing result indicated that the A allele had C-T mutation at the CDS partial sequence in 52 bp.The correlation analysis showed that the wool fineness had no significant difference among different genotypes(P〉0.05 ).
