丁香酚减轻蛛网膜下腔出血大鼠脑血管痉挛 被引量:2

The effect of Eugenol on the cerebral vasospasm in a rat subarachnoid hemorrhage model

摘要 目的观察丁香酚对蛛网膜下腔出血(subarachnoid hemorrhage,SAH)大鼠脑血管痉挛(cerebral vasospasm,CVS)的治疗作用。方法血管内插线法建立大鼠SAH模型后,立即吸入丁香酚,每天3次,共7天。测量各组基底动脉(basilar artery,BA)的直径和管壁厚度,ELISA方法测量各组BA内MPO和MDA含量,免疫印迹法检测BA的p-p53、caspase-3、NF-κB和TNF-α的表达。结果丁香酚可增加蛛网膜下腔出血大鼠BA直径并减小管壁厚度,显著减少BA内MPO和MDA含量,下调p-p53、caspase-3、NF-κB和TNF-α表达水平。结论丁香酚减轻脑血管痉挛可能与抑制BA内的细胞凋亡和炎症反应相关。 Objective To explore the protective role of Eugenol in attenuating the severity of cerebral vasospasm(CVS) after subarachnoid hemorrhage(SAH). Methods The SAH model was established in male SD rats by using endovascular perforation method. After SAH, the Eugenol was administrated by olfactory inhalation 30 min, three times per day. At 7 days following SAH, the diameter and wall thickness of basilar artery(BA) were measured after performing HE staining, the levels of myeloperoxidase(MPO) and malondialdehyde(MDA) in BA were evaluated by ELISA methods. The levels of p-p53, cleaved-caspase-3, NF-κB and TNF-α in BA were determined by Western blot. Results After Eugenol treatment, the diameter of BA was markedly increased and the thickness of vessel wall was significantly decreased, additionally, the contents of MPO and MDA and the expression levels of p-p53, cleaved-caspase-3, NF-κB(p65) and TNF-α in BA were significantly reduced. Conclusion The alleviation of CVS severity of SAH by Eugenol might be related to blocking apoptosis and inflammation pathway in the cerebral vessels.
