摘要
目的通过猪体免疫试验,评价欧洲型PRRSV重组痘苗病毒灭活疫苗的免疫效果。方法分别用重组痘苗病毒rVV-EU-ORF3-ORF5和灭活rVV-EU-ORF3-ORF5进行猪体免疫试验,通过检测猪血清特异性抗体中和抗体检测及T淋巴细胞增殖试验分析免疫效果。实验设E3L-VTT野毒免疫组和PBS组作对照。结果免疫后35d,欧洲型PRRSV重组痘苗病毒灭活疫苗免疫组和rVV-EU-ORF3-ORF5免疫组GP3、GP5特异抗体水平均达峰值(A值0.51~0.56),两组间比较差异无统计学意义(tGP3=1.805,tGP5=0.7998,P〉0.05),但均高于E3L-VTT野毒免疫组(A值0.33~0.43),差异显著(tGP3=5.518,tGP5=3.346,P〈0.01);猪血清中和抗体重组痘苗病毒灭活疫苗组与重组痘苗病毒疫苗组均与免疫后35d达峰值水平(分别为16.38和18.88),且两组间比较差异无统计学意义(t=0.7005,P〉0.05);T淋巴细胞增殖试验的SI值两疫苗组差异无统计学意义(t=0.2536,P〉0.05),但均高于E3L-VTT野毒免疫组(t值分比为6.414和7.222,P〈0.01)。结论欧洲型PRRSV重组痘苗病毒灭活疫苗能诱导猪体产生体液和细胞免疫应答,可为欧洲型PRRSV的新型疫苗的研制提供参考。
Objective The immunological properties of an inactivated recombinant vaccinia vaccine against the European porcine reproductive and respiratory syndrome virus(PRRSV)were evaluated in an immunization experiment involving pigs. Methods Pigs free of the European PRRSV were immunized with the recombinant vaccinia vaccine rVV-EUORF3-ORF5 or the inactivated recombinant vaccinia vaccine rVV-EU-ORF3-ORF5,and the level of virus-specific neutralizing antibodies was determined,T-lymphocyte proliferation was assayed,and the GP3 and GP5proteins of the European PRRSV were detected.Pigs administered E3L-VTT or PBS served as control groups. Results The levels of GP3 and GP5-specific antibodies peaked in both pigs given the recombinant vaccinia vaccine and pigs given the inactivated recombinant vaccinia vaccine 35 days post-immunization(A=0.51~0.56).Differences in antibody levels did not differ significantly between pigs given the recombinant vaccinia vaccine and pigs given the inactivated recombinant vaccinia vaccine(tGP3=1.805,tGP5=0.7998,P〉0.05).However,antibody levels were markedly higher in the immunized groups than in pigs administered E3L-VTT(A=0.33~0.43),and the difference in antibody levels was statistically significant(tGP3=5.518,tGP5=3.346,P〈0.01).Detection of neutralizing antibodies in serum indicated that pigs immunized with the inactivated recombinant vaccinia vaccine or the recombinant vaccinia vaccine had high levels of antibodies to the GP3 and GP5proteins of the European PRRSV,and these levels peaked 35 days post-immunization(16.38 and 18.88,respectively).However,the difference in antibody levels between the 2groups was not statistically significant(t=0.7005,P〉0.05).Results of a T-lymphocyte proliferation assay indicated that SI did not differ significantly in pigs given the recombinant vaccinia vaccine and pigs given the inactivated recombinant vaccinia vaccine(t=0.2536,P〉0.05).SI did differ significantly in pigs administered E3L-VTT(t=6.414,7.222,P〈0.01).
