川芎-天麻治疗偏头痛肝阳上亢证大鼠的作用机制研究 被引量:10

Therapeutic Mechanism of Drug Pair of Rhizoma Chuanxiong and Rhizoma Gastrodiae for Migraine Rats with Hyperactivity of Liver-yang

摘要 目的比较川芎-天麻不同比例水提物、醇提物对偏头痛肝阳上亢证大鼠的疗效,并探讨其可能的作用机制。方法取昆明种小鼠80只,雌雄各半,随机分为8组,每组10只,即正常组,模型组,川芎-天麻(4∶1,1∶1,1∶4)水提组、川芎-天麻(4∶1,1∶1,1∶4)醇提组。川芎-天麻水提组和醇提组均按1.75 g·kg-1灌胃给予相应提取液1次,正常组和模型组给予等量清水。小鼠腹腔注射0.6%醋酸溶液,记录15 min内小鼠扭体次数。以醋酸扭体法评价川芎-天麻对小鼠的镇痛作用。取SD大鼠64只,随机分为8组,每组8只,即正常组,模型组,川芎-天麻(4∶1,1∶1,1∶4)水提物组、川芎-天麻(4∶1,1∶1,1∶4)醇提物组。用附子和硝酸甘油复制偏头痛肝阳上亢证大鼠模型,测定血清一氧化氮合酶(NOS)活性,大鼠降钙素基因相关肽(CGRP)、一氧化氮(NO)、内皮素(ET)、血栓素B2(TXB2)、6-酮-前列腺素1α(6-Keto-PGF1α)含量,并计算NO/ET、TXB2/6-Keto-PGF1α比值。结果川芎-天麻不同比例醇提物和水提物均能降低醋酸诱发的小鼠扭体次数,与模型组比较,差异有统计学意义(P<0.05)。与同比例水提组比较,川芎天麻(4∶1)醇提组可降低模型组CGRP、NOS活性、NO含量、NO/ET,(1∶1)醇提组降低模型组NOS活性、NO/ET、TXB2/6-Keto-PGF1α比值,升高6-Keto-PGF1α含量,(1∶4)醇提组降低模型组CGRP、NOS活性、NO含量、NO/ET、TXB2/6-Keto-PGF1α比值,升高6-Keto-PGF1α含量,差异有统计学意义(P<0.05,P<0.01)。结论川芎-天麻醇提物组治疗偏头痛效果优于水提物组,其调节模型大鼠血清CGRP、NOS、NO、ET、TXB2、6-Keto-PGF1α等指标的水平及NO/ET、TXB2/6-Keto-PGF1α平衡,可能是其治疗偏头痛的部分机制。 Objective To compare the effect of water extract and alcohol extract of drug pair of Rhizoma Chuanxiong (Chuanxiong) and Rhizoma Gastrodiae(Tianma) in different combination proportions on migraine rats with hyperactivity of liver-yang, and to investigate the possible mechanism. Methods Eighty Kunming mice with half being male were evenly randomized into 8 groups, namely normal group, model group, Chuanxiong-Tianma(in the proportion of 4 : 1, 1 : 1, 1 : 4, respectively) water extract and alcohol extract groups. Chuanxiong-Tianma water extract and alcohol extract groups were given gastric gavage of the corresponding extract 1.75 g·kg-1 once a day, and the normal group and model group were given the same volume of water. The mice were injected with 0.6 % acetic acid solution intraperitoneally, and then we recorded the mice body torsion frequency within 15 min to evaluate the analgesic effect of Chuanxiong-Tianma. Sixty-four SD rats were also evenly randomized into 8 groups like the Kunming mice, and the model of hyperactivity of liver-yang was induced by nitroglycerin and aconite decoction. After treatment, we tested the serum nitric oxide synthase (NOS) activities, and serum contents of calcitonin-gene-related peptide (CGRP), nitric oxide(NO), endothelin(ET), thromboxane B2(TXB2), and 6-keto- prostaglandin 1 α (6-Keto-PGF1α ), and then we counted the ratio of NO/ET and TXB2/6-Keto-PGF1 α in the rat serum. Results Chuanxiong-Tianma water extract and alcohol extract in different proportions all decreased the mice body torsion frequency within 15 rain, the difference being significant compared with that in the model group(P 〈 0.05). Compared with the water extract groups in the same proprotion, Chuanxiong-Tianma(4 : 1 ) alcohol extract group had better effect on reducing CGRP, NOS, NO, and NO/ET; Chuanxiong-Tianma( 1 : 1 ) alcohol extract group had better effect on reducing NOS, NO/ET, TXB2/6-Keto- PGF1α and on increasing 6-Keto-PGF1α; Chuanxiong-Tianma
