siRNA干扰蛋白激酶D2表达抑制结直肠癌细胞的侵袭转移 被引量:1

摘要 目的:观察蛋白激酶D2(PKD2)对结直肠癌细胞侵袭转移作用的影响。方法:采用免疫印迹及免疫组织化学染色观察结直肠癌细胞及组织中PKD2的表达;免疫印迹观察PKD2对上皮间质转化(EMT)相关分子表达水平的影响;克隆形成实验、划痕实验和侵袭小室观察PKD2对结直肠癌细胞成瘤、迁移及侵袭作用的影响。结果:PKD2在结直肠癌中表达升高,且晚期结直肠癌表达较早期结直肠癌表达增高(P<0.05);siRNA干扰PKD2的表达可抑制结直肠癌细胞的EMT;siRNA干扰PKD2表达抑制结直肠癌细胞的成瘤、迁移及侵袭作用。结论:在结直肠癌中,抑制PKD2的表达可抑制肿瘤细胞的侵袭转移,PKD2的表达与结直肠癌的进展呈正相关。 Objective: To investigate the role of protein kinase D2 in CRC cells invasion and migration. Methods: PKD2 expression was detected by western blotting or Immunohistochemistry in both CRC cells and CRC tissues. Epithelial-mesenchymal transition related markers were detected by western blotting between control siRNA and PKD2 siRNA. Clone soft agar,scratches and transwell were performed to investigate the influence of PKD2 to the tumorgenesis,invasion and migration of colorectal cancer cells. Results: PKD2 protein was high expressed in the CRC cells,especially in advanced carcinoma cells( P〈 0. 05). Inhibition of PKD2 by siRNA decreased EMT markers in CRC cells. PKD2 reduction inhibited tumorgenesis,invasion and migration of CRC cells. Conclusion: PKD2 reduction by siRNA inhibits the invasion and migration of CRC. The expression of PKD2 positively relates with CRC progression.
