FSH和LH对滩羊卵母细胞体外成熟培养的影响 被引量:2

摘要 为了探讨滩羊卵母细胞体外成熟过程中促卵泡素(FSH)和促黄体素(LH)的最佳添加量,试验设为4组:对照组、FSH组(10、50、100、200、300μg/m L 5个质量浓度梯度)、LH组(5、10、20、50、100μg/m L 5个梯度)和FSH&LH组(100μg/m L FSH+20μg/m L LH),对滩羊卵母细胞的体外成熟培养进行研究,统计第一极体排出率.结果表明:100μg/m L FSH+20μg/m L LH组卵母细胞成熟率最高,为64.64%,明显高于其他3组,差异显著(P<0.05);FSH组中100μg/m L优于其他4个质量浓度梯度和对照组,差异显著(P<0.05);LH组的20μg/m L的质量浓度优于其他4组和对照组,差异显著(P<0.05).因此,在本试验条件下,100μg/m L FSH+20μg/m L LH是滩羊卵母细胞体外成熟培养的最佳培养添加质量浓度. To investigate the appropriate concentration of FSH and LH in TAN sheep oocyte maturation in vitro, TAN sheep oocytes were divided into four groups in vitro maturation culture: the control group, the FSH group, the LH group and the FSH&LH group. In the FSH group, the concentration of FSH was 10μg/ml, 50μg/ml, 100μg/ml, 200μg/ml and 300 μg/ml respectively, in the LH group, the concentration of LH was 5μg/ml, 10μg/ml, 20μg/ml, 50μg/ml and 100 μg/ml respectively, and the concentration of FSH and LH were respectively added 100 μg/ml and 20μg/ml in the FSH&LH group. The rates of polar body I (Pb I) expulsion was analyzed in the test. The results showed that the rate of oocytes maturation of the FSH&LH group was 64.64%, which was the highest rate in each group (P〈0.05), the rate of oocytes maturation of the FSH&LH group was 64.64%, which the highest rate in each group (P〈0.05), the FSH group including 100 μg/ml is better than the other four concentration gradient and the control group (P〈0.05), the LH group including 20μg/ml was better than the other four concentration gradient and the control group (P〈0.05). Therefore, adding 100 μg/ml FSH and 20 μg/ml LH was the appropriate concentration of FSH and LH in TAN sheep oocyte maturation in vitro under this experimental condition.
