摘要
斑茅是栽培种甘蔗最重要的近缘物种之一,具有抗逆性强等特点。本文以高粱过氧化氢酶基因(NCBI登录号为XM002437586.1)c DNA序列为探针,对甘蔗属EST数据库进行同源检索、筛选和拼接,后经基因组PCR、分子克隆、序列分析验证成功分离了1个斑茅过氧化氢酶基因DNA序列(Ea CAT-1a,Gen Bank登录号为KF864223)。该序列全长3831 bp,包含8个外显子和7个内含子,c DNA序列长为1532 bp,包含10 bp的5’非翻译区(UTR)和43 bp的3’UTR,以及一个1479 bp的开放读码框,编码492个氨基酸,此蛋白序列具有过氧化氢酶保守结构域,属于CAT家族。将推测的Ea CAT-1a蛋白序列与高粱、水稻、玉米等物种进行同源进化分析,均表现出较高的保守性。
Erianthus arundinaceus( Retz.) Jesw. is one of the most important species of sugarcane related species,and with strong resistance. In this paper,one novel full-length catalase gene( Ea CAT-1a,Gen Bank Accession number KF864223) DNA sequence was cloned by blasting the Saccharum L. EST database using sorghum CAT gene( Gen Bank Accession number XM 002437586. 1) c DNA sequence as a querying probe. Ea CAT-1a was 3831 bp,containing 8 exons and 7 introns,length of c DNA was 1532 bp,containing 10 bp of 5 'untranslated region( UTR) and 43 bp of the 3'UTR,and a 1479 bp open reading frame encoding a 492 amino acid sequence of the protein which has a conserved domain of catalase belonging CAT family. The putative Ea CAT-1a protein sequence was very homologous with sorghum,rice,maize and other species,and have shown a high conservatism by homologous evolutionary analysis.
