摘要
目的体外观察5-氟尿嘧啶(5-FU)与姜黄素联用对人结肠癌LoVo细胞的生长抑制和凋亡诱导的作用。方法采用MTT方法、流式细胞检测技术和荧光染料AO/EB双染法观察不同浓度姜黄素和5-氟尿嘧啶单独或联合应用对Lovo细胞生长和凋亡的影响。结果 MTT法检测结果显示,各药物组LoVo细胞的生长均受到了显著抑制,各剂量5-FU以及分别联合姜黄素对LoVo细胞的抑制率呈剂量-时间依赖关系;FCM检测结果表明,姜黄素联合低剂量5-FU组的早期、晚期凋亡率显著高于中剂量和高剂量5-FU单用组,姜黄素联合中剂量5-FU组的细胞凋亡率最高;荧光显微镜观察发现各实验组细胞同对照组相比呈现典型凋亡形态,且联合用药组细胞凋亡率最高。结论姜黄素与低剂量5-FU联用对体外培养的结肠癌LoVo细胞的的生长抑制和促凋亡作用能够达到高剂量5-FU单用的效果。
Objective To observe the growth inhibitory effect of Curcumin combined with fluorouracil( 5-FU) on the colorectal cancer cell line LoVo in vitro. Methods By adopting the MTT,Flow Cytometer,acridine orange( AO) / ethidium bromide( EB)doubly staining method,the growth inhibition and apoptosis induction effect of different concentration of Curcumin and 5-fluorouracil alone or in combination on the human colon carcinoma LoVo cells are tested. Results MTT assay showed that each group of LoVo cell growth was inhibited,each dose of 5-FU with a certain amount of Curcumin joint action in inhibitory rate of LoVo cells are shown as a dose-dependent. The results of FCM showed that,the early and late-stage apoptosis rate of the group of Curcumin combined with low dose of 5-FU were significantly higher than the groups of moderate and high dose alone,and the group of Curcumin combined with moderate dose 5-FU has the highest rate of apoptosis; With fluorescence microscope,we found that,each of the experimental group compared with the control group renders the typical morphology of apoptosis,and the combination group has the highest rate of cell apoptosis. Conclusion Curcumin with low concentrations of fluorouracil can achieve the effect of high dose 5-FU alone in proliferation of LoVo cells and induce apoptosis.
