淮山药叶乙醇提取物黄酮和皂苷含量及抗氧化抗肿瘤活性研究 被引量:9

Antioxidant and cytotoxic activity of ethanol extract of Rhizoma Dioscoreae leaves

摘要 研究淮山药叶乙醇提取物的黄酮和皂苷含量,抗氧化活性及其对肿瘤细胞A549、MCF-7,SW480与人脐静脉血管内皮细胞的抑制作用。结果发现,淮山药叶乙醇提取物中含有大量的黄酮和皂苷类物质,含量分别为(443.27±21.15)mg芦丁当量/g,(362.73±28.83)mg薯蓣皂苷当量/g(以干重计);DPPH自由基清除活性接近于抗坏血酸;24 h细胞培养试验表明提取物对肿瘤细胞的抑制作用呈明显的量效关系,而对正常细胞的抑制作用浓度超过200μg/m L。由此可见,淮山药叶是一种有益于健康的物质来源。 The aim of this study was to evaluate chemical composition, antioxidant and cytotoxic effect of ethanol extract of Rhizoma Dioscoreae leaves on lung adenocarcinoma (A549), breast cancer (MCF- 7), colorectal adenocarcinoma (SW480) and human umbilical vein endothelial (HUVEC) cell lines. High flavonoid and saponin content (443.27±21.15) mg RE/g of dry weight (DW), (362.73±28.83) mg DE/g of DW, respectively) in the ethanol extract was found by spectrophotometric methods. The ethanol extract of Rhizoma Dioscoreae leaves displayed significant antioxidant and cytotoxic activity. Its DPPH radical scavenging activity was close to ascorbic acid (Vc). Dose-dependent inhibition of growth of cancer cell lines after 24 h treatment was observed while normal cell (HUVEC) growth was inhibited above a culture concentration of 200 μg/mL extract. These revealed that Rhizoma Dioscoreae leaves are good sources of supplements for human health.
