摘要
目的研究三七总皂苷(panax notoginseng saponin,PNS)抗心肌细胞肥大是否与成纤维细胞生长因子诱导早期反应蛋白14(fibroblast growth factor-inducible 14,Fnl4)的表达变化相关。方法分离原代新生大鼠心肌细胞,去甲肾上腺素(Norepinephrine,NE)处理原代细胞建立心肌细胞肥大模型。鬼笔环肽染色观察细胞形态,实时定量聚合酶链反应(quantitative real-time polymerase chain reaction,q RT-PCR)检测肥大相关基因和Fn14 m RNA表达水平,Western blot检测Fn14蛋白表达水平。结果与对照组比较,NE处理组细胞表面积增大、细胞肥大相关基因m RNA表达水平明显升高,Fn14 m RNA和蛋白表达升高,差异均有统计学意义(P<0.05)。与单独NE组比较,NE和PNS同时处理组的细胞表面积减小、肥大相关基因m RNA表达水平显著下降,差异有统计学意义(P<0.05)。PNS对Fn14 m RNA表达水平影响不大,但是却明显的促进Fn14蛋白表达(P<0.05)。结论 PNS能够抗NE诱导的心肌细胞肥大,同时能够影响Fn14的表达。PNS抗心肌肥大可能通过Fn14起作用。
Objectives To investigate whether inhibition effect of panax notoginseng saponin(PNS) on cardiac hypertrophy is related to fibroblast growth factor-inducible 14(Fn14) expression. Methods The cardiomyocytes were separated from the neonatal rat hearts. Norepinephrine(NE) was used as an inducer of cardiac hypertrophy. Phalloidin was used to indicate cell morphology. The m RNA expression of hypertrophic genes and Fn14 were detected by quantitative real-time polymerase chain reaction(q RT-PCR). Western blot assay was used to detect the protein expression of Fn14. Results The cell surface increased and m RNA expression of hypertrophic genes was obviously upregulated by NE compared with that in controls(P〈0.05); Both the m RNA and protein expressions of Fn14 were also upregulated by NE(P〈0.05). PNS decreased the cells surface and downregulated the hypertrophic gene expression induced by NE. Though it had little effect on the m RNA expression of Fn14, PNS obviously upregulated the protein expression of Fn14(P〈0.05). Conclusions PNS can inhibit cardiac hypertrophy induced by NE and upregulate Fn14 protein expression, with Fn14 as a pathway.
