微环载体与基因治疗

摘要 基因治疗是将有治疗作用或正常基因的DNA序列导入靶细胞,通过纠正基因缺陷或发挥治疗作用而达到治疗疾病的目的。基因治疗的关键技术之一是研发用于真核细胞转移基因的安全有效的载体系统。病毒和非病毒载体系统均有各自的优势和缺点。微环载体是一种来源于质粒DNA的新型的较小的超螺旋表达元件,通过在大肠杆菌体内发生位点特异性结合而产生,并已应用于基因治疗。由于微环载体缺乏原核DNA骨架序列,如抗生素基因、原核复制起始点等,可提高治疗基因表达水平、基因治疗的安全性及降低炎症反应。本文就微环载体的产生、提高转基因表达的机制及在基因治疗中的应用作一综述。
