摘要
目的观察缺氧诱导因子-1α(hypoxia-inducible factor-1α,HIF-1α)、胰岛素样生长因子Ⅱm RNA结合蛋白3(insulin-like growth factor-Ⅱm RNA-binding protein 3,IBMP3)、增殖细胞核抗原(proliferating cellnuclear antigen,PCNA)和P53蛋白在反流性食管炎(ruflex esophagitis,RE)、barrett食管(Barrette esophagus,BE)、食管腺癌(esophageal edenocarcinoma,EAC)组织中的表达情况,分析HIF-1α、IMP3、PCNA和P53的表达在EAC发生过程中的意义。方法采用免疫组化法,检测50份RE、100份BE和100份EAC样本中HIF-1α、IMP3、PCNA和P53蛋白的表达,并取无反酸、烧心等症状,无免疫疾病、感染疾病病史及胃镜下食管下段黏膜无明显病变的功能性消化不良患者样本30份作为对照。结果 HIF-1α蛋白在RE、BE和EAC组中阳性率分别为14%、54%和92%,与对照组相比,差异有统计学意义(P均<0.05);P53蛋白在RE、BE和EAC组中阳性率分别为8%、31%和84%,EAC和BE组与对照组相比,差异有统计学意义(P均<0.01);IMP3蛋白在RE、BE和EAC组中阳性率分别为10%、45%和74%,EAC和BE组与对照组相比,差异有统计学意义(P均<0.01);PCNA蛋白在RE、BE和EAC组中阳性率分别为22%、47%和74%,EAC和BE组与对照组相比,差异有统计学意义(P均<0.01);EAC与BE组及BE与RE组比较,HIF-1α、P53、IMP3、PCNA蛋白的表达差异有统计学意义(P均<0.01),4种蛋白在RE、BE和EAC组中的表达逐渐增强。HIF-1α表达与IMP3、P53、PCNA表达均呈正相关(P<0.01)。结论 HIF-1α、IMP3、PCNA和P53蛋白的表达与BE、EAC的发生发展密切相关,可用作监测BE、EAC早期发生的指标,为今后BE及EAC的治疗提供新的途径。
Objective To observe the expressions of hypoxia-inducible factor-1α(HIF-1α), insulin-like growth factor-Ⅱm RNA-binding protein 3(IMP3), proliferating cello nuclear antigen(PCNA)and P53 protein in ruflex esophagitis(RE),Barrett esophagus(BE) and esophageal edenocarcinoma(EAC) tissues, and analyze their significance in pathogenesis of EAC. Methods The expressions of HIF-1α, IMP3, PCNA and P53 in 50 samples of RE, 100 samples of BE and 100 samples of EAC were determined by immunohistochemical assay with SP staining, using those in 30 samples from patients with functional dyspepsia, without symptoms of acid regurgitation and heartburn, histories of immune or infectious diseases or significantly pathological change in esophageal mucosa under gastroscope, as control. Results The positive rates of HIF-1α in RE, BE and EAC groups were 14%, 54% and 92% respectively, which showed significant difference with that in control group(each P 〈 0. 05). The positive rates of P53 in RE, BE and EAC groups were 8%, 31% and 84%respectively, and the two latter showed significant difference with that in control group(each P 〈 0. 01). The expression of HIF-1α was positively related to those of IMP3, P53 and PCNA(each P 〈 0. 01).Conclusion The expressions of HIF-1α, IMP3, PCNA and P53 were closely related to the pathogenesis and progress of BE and EAC, which might be served
