化浊行血颗粒对动脉粥样硬化模型大鼠颈总动脉PPARs的影响 被引量:1

The Influence of Huazhuo Xingxue Particles on General Carotid PPARs of Model Rats with Atherosclerosis

摘要 目的:研究化浊行血颗粒对食饵性动脉粥样硬化(atherosclerosis,AS)模型大鼠颈总动脉过氧化物酶体增殖物激活受体(peroxisome proliferator-activated receptor,PPARs)的影响,探讨中药治疗AS的有效方法。方法:实验动物按体质量随机分为正常对照组、模型组和药物组,每组12只,采用高脂饲料法建立大鼠食饵性AS模型。建模后,药物组灌胃给予化浊行血颗粒水溶液[12 g生药/(kg·d)-1],正常对照组、模型组分别给予等容量的蒸馏水,连续8周。实时PCR(real time PCR,RT-PCR)法测定PPARmRNA。结果:与正常对照组比较,模型组颈动脉内PPARα、PPARβ及PPARγ含量显著降低(P<0.01);药物组PPARα、PPARβ及PPARγ含量显著升高(P<0.01)。药物组与模型组比较,PPARα、PPARβ及PPARγ含量显著升高(P<0.01)。结论:PPARs可抑制AS形成,化浊行血颗粒可有效激活PPARα、PPARβ及PPARγ,从而抑制AS的发生、发展。 Objective: To study the influence Hnazhuo Xingxue Particles (HZXXP) on General earotid peroxisome proliferator-aetivated rcceptu (PPARs) of model rats with atherosclerosis ( AS), and explore the effective TCM therapies to treat AS. Methods: The experimental rats were randomly divided into the normal control group, the model group and the medicine group, with 12 cases in each gronp. The rats prey AS models were established by high fat diet method. Then the ones in the medicine group were given HZXXP salutioo by garage ( 12g raw/kg/d) , while the ones in the normal control group and the model group were given than same amount of distilled water respectively for 8 weeks continuously. PPARmRNA was detected by using PCR( real time PCR, RT-PCR). Results: Compared with the normal control group, PPARα, PPARβ and PPARγeontents in carotid of the model group decreased obviously ( P 〈 0.01 ) , while those of the medicine group increased obviously ( P 〈 0.01 ) ; Compared with the model grot,p, the above indexes of the medicine group increased obviously (P 〈 0.01 ). Conclusion: PPARs can impress the formation of AS, and HZXXP can effectively activate PPARα, PPARβ and PPARγ e to control the occurrence and development of AS.
