摘要
[目的]探讨微波法提取广西凌云特产新鲜川木瓜(皱皮木瓜)总酚的最优工艺。[方法]以没食子酸为对照品,用紫外-可见分光光度法,测定广西凌云特产川木瓜总酚含量。采用正交试验纤维素酶辅助提取的方法,考察提取溶剂浓度、微波功率、提取时间、提取次数、料液比等因素对广西凌云特产川木瓜总酚提取率的影响。[结果]以乙醇浓度80%,微波输出功率中高火,微波处理时间90 s,提取2次,料液比1∶3 g/ml 5个因素水平为最佳提取条件,此条件下凌云特产川木瓜总酚的提取率为0.044 3%。[结论]此方法操作简便,重复性、稳定性良好,准确可靠,可为广西凌云特产川木瓜的质量控制及开发利用提供一定的方法和依据。
[ Objective ] To explore the optimal process method of microwave extraction for the total phenol in Guangxi Lingyun specialty Chuan papaya( Chaenomeles speciosa Nakal). [ Method ] With garlic acid as reference substance, using UV-VIS spectrophotometry, total phenol con- tent in Guangxi Lingyun specialty Chuan papaya(Chaenomeles speciosa Nakal) was determined. Use orthogonal test method with Cellulase as- sisted to investigate following factors as extraction solvent, microwave power, extraction time, liquid ratio and extraction times, on total phenol in the Guangxi Lingyun specialty Chuan papaya( Chaenomeles speciosa Nakal). [ Result] In 80% ethanol concentration, microwave output power on high fire, microwave heating time of 90 s, extracted twice, and solid-liquid ratio 1 : 3 g/ml for these five factors is the best. On this condition, extraction rate of total phenol that Lingyun specialties Chuan papaya (Chaenomeles speciosa Nakal )yield of total phenol was 0. 044 3%. [ Conclusion] The results show that this method is simple, good repeatability, stability, accurate and reliable. It provides some methods and basis for the development and utilization for quality control of Guangxi Lingyun specialty Chuan papaya (Chaenomeles soeciosa Nakal).
