摘要
以红网纹草的顶芽、叶片、茎段为外植体,对其进行最佳消毒方法的研究。首先以叶片为外植体,筛选最适宜的次氯酸钠浓度和升汞浓度,即进行不同浓度次氯酸钠(1%,2%,5%)和升汞(0.05%,0.10%,0.15%)的单因素试验。继而进行外植体(顶芽,叶片,茎段)、2%次氯酸钠消毒时间(0,5,10 min)、0.10%升汞消毒时间(0,5,10 min)3因素3水平的L9(34)正交试验。结果表明:在单因素试验中,适合红网纹草外植体的次氯酸钠和升汞浓度分别为2%和0.10%;在正交试验中,茎段与叶片污染率相同,均低于顶芽,但茎段成活率最高,因而为理想的外植体,最佳的消毒方法是将茎段外植体在2%的次氯酸钠中消毒5 min,再在0.10%的升汞中消毒5 min。此种消毒方法污染率和死亡率最低为0,存活率最高为100%,是适宜红网纹草的最佳消毒方式。
Taking Fittonia verschqffeltii terminal buds, leaves, and stem segments as explants, the experiment was carried out to find out the best disinfention method. Firstly, with leaves as its explants, the single factor experiment was carried out with three concentrations of NaCIO (2%, 5%,10%) and HgCl2(0.05%, 0.10%, 0.15%). Secondly, an orthogonal experiment followed with L9 (34)array of three factors and three levels, namely explants(terminal bud, leaf, stem segment), the disinfection time for NaC10(0,5,10 min), and the disinfection time for HgC12 (0,5,10 min).The results showed that in the single factor experiment with leaf as explants, the concentrations of NaCIO and HgC12 with good disinfection effect were 2% and 0.10%respectively. In the orthogonal test, stem segments, compared with terminal buds and leaves, had low pollution rate and high survival rate, and were easier to produce callus, leading to its becoming the ideal explants for the tissue culture of Fittonia verschaffeltii. The best disinfection way was that the explants were immersed in alcohol(75%) for 30 s, NaC10(2%) for 5 rain and then HgC12(0. 10%) for 5 rain with the lowest pollution rate(O%) and highest survival rate(100%).
