摘要
目的:探讨大黄素对于肝硬化门脉高压大鼠肠屏障有无保护作用。方法:SD雄性大鼠25只,随机分为对照组(A组)5只、肝硬化模型组(B组)10只、大黄素干预组(C组)10只,500 m L/L四氯化碳(3 m L/100 g)皮下注射8周复制肝硬化大鼠模型,C组在制作模型第15天起同时每日大黄素(5 mg/m L,5 m L/kg)灌胃,1/d,A组大鼠给予生理盐水(3 m L/100 g)皮下注射,A组和B组每日给予相当体积的生理盐水灌胃,8周后处死大鼠,行墨汁推进试验测定肠道传输功能,测定门脉压力,取末端回肠组织观察组织病理学改变、血浆内毒素水平测定。结果:(1)B组小肠黏膜损伤评分明显高于A组(P值均<0.05),C组小肠黏膜损伤评分较B组减轻。(2)B组门脉压力与A组比较,门脉压力显著升高,P<0.05,大鼠明显处于高循环状态;C组门脉压力与B组比较明显降低,P<0.05。(3)B组血浆内毒素水平与A组比较,血浆内毒素水平显著升高,P<0.05,C组血浆内毒素水平与B组比较明显降低,P<0.05;(4)B组肠道黑染百分比与A组比较明显缩短,P<0.05,C组肠道黑染百分比与B组比较明显延长,P<0.05。结论:应用大黄素干预大鼠肝硬化进程后,大黄素组大鼠肝脏胶原纤维与网状纤维沉积较少,假小叶少见。大鼠的门脉压力有明显的下降,大黄素组大鼠血浆内毒素水平明显下降,光镜下小肠黏膜损伤评分显著降低,肠动力明显好转,大黄素对于肝硬化门脉高压大鼠的肠屏障功能有保护作用。
Objective : To investigate the effects of emodin on intestinal mncosal barrier in cirrhotic rat with portal hypertension. Methods :Twentyfive male SD rats were randomly divided into three groups:control group( n = 5 ), model group( n = 10) ,emodin group (n = 10). The model was induced by the subcutaneous injection of 500 mL/L CCL4 (3 mL/100g) ( twice a week for 8 weeks ), ( 5mg/mL, 5mL/kg) emodin was intragastrically administrated in emodin group from the fifteenth day, once a day while the control group received normal saline instead. The same volume saline was in- tragastrically administrated in control and model groups. The changes of Ink propelling test, Portal pressure, Histology, En- dotoxin in those rats were observed. Results: The degrees of intestinal mucosal injury, Portal pressure and Endotoxin in model group were higher than those of the control group while the degrees of intestinal mucosal injury, portal pressure and Endotoxin in emodin group were lower than those of the model group. The percentage of dyed intestinal black in emodin group was higher than that of the model group. Conclusion Emodin can alleviate intestinal pathological damages, reduce portal pressure and improve intestinal motility. The emodin could protect intestinal mucosal barrier in cirrhotic rats with portal hypertension.
