摘要
目的研究氯喹(chloroquine,CQ)对杨梅素诱导的He La细胞凋亡的影响。方法体外培养宫颈癌He La细胞,实验分为Control组、单加杨梅素(50μg/ml)组、单加CQ(20μM/ml)组、合加药物组(杨梅素50μg/ml+CQ 20μg/ml)共4组,观察药物对He La细胞增殖率的影响。共聚焦显微镜下观察He La细胞核的形态变化、凋亡相关蛋白及自噬标志物的表达。结果与单加杨梅素组相比,合加药物组He La细胞增殖率明显下降,凋亡形态学改变更加明显及凋亡相关蛋白active caspase-3表达增加;自噬标志物微管相关蛋白轻链3(microtubule-associated protein light chain 3,LC3)表达增加。结论氯喹能够促进杨梅素诱导的HeLa细胞凋亡。
Objective To research the effect of chloroquine on myricetin induced apoptosis in He La cells. Methods Human cervical cancer He La cells were cultured in vitro. In order to observe the drug effect on proliferation rate of He La cells,He La cells were divided into 4 groups: control group,myricetin group( 50 μg / ml),chloroquine group( 20 μM / ml),combination group( myricetin 50 μg / ml +chloroquine 20 μM / ml). Confocal microscopy was used to detect morphological changes of He La cell nucleus,expressions of apoptosis related protein and autophagy markers. Results Compared with myricetin group,proliferation rate of He La cells in combination group significantly decreased with more obvious morphological changes and higher apoptosis related protein active caspase- 3 expression; the expression of autophagy marker microtubule- associated protein light chain 3( LC3) increased. Conclusion Chloroquine promotes myricetin induced apoptosis in He La cells.
