摘要
目的探讨人肝癌细胞系Hep G2中乙型肝炎病毒表面抗原(HBs Ag)对该细胞增殖及I型胶原(Col I)、α-平滑肌肌动蛋白(α-SMA)分泌的影响。方法采用含不同浓度HBs Ag的培养基对Hep G2细胞组(A组)、LX-2和Hep G2共培养组(B组)细胞进行培养,CCK-8法观察不同浓度的HBs Ag对两组Hep G2细胞增殖的影响,并设含普通培养基的Hep G2组(C组)和LX-2和Hep G2共培养组(D组)进行比较;ELISA法观察敏感浓度的HBs Ag对各组Hep G2细胞Col I、α-SMA分泌的影响。结果随着HBs Ag浓度的升高,各组Hep G2细胞增殖率呈上升趋势,且浓度5 ng/ml时增殖率明显升高,差异有统计学意义(P<0.05)。采用5 ng/ml浓度HBs Ag刺激48 h后,与C组比较,A组细胞上清液中Col I和α-SMA水平明显升高,差异有统计学意义(t=3.419,P=0.005;t=4.388,P=0.001);与D组比较,B组Col I和α-SMA表达水平明显升高,差异有统计学意义(t=3.048,P=0.009;t=3.005,P=0.009)。结论 HBs Ag可以促进人肝癌细胞Hep G2的增殖;Hep G2细胞具有分泌Col I和ɑ-SMA的类似肝星状细胞的特质,HBs Ag可能通过刺激肝癌细胞Hep G2和肝星状细胞LX-2共同促进Col I和ɑ-SMA的分泌而促进肝纤维化、肝硬化和肝癌的进展。
Objective To investigate the effect of hepatitis B virus surface antigen( HBs Ag) on cell proliferation and secretion of type I collagen( Col I),ɑ-smooth muscle actin( ɑ-SMA) in human hepatocellular carcinoma line Hep G2. Methods Hep G2 cell group( group A),LX-2 and Hep G2 trained group( group B) were cultured by cell supernatants which contained different concentrations of HBs Ag. The effects of different HBs Ag on cell proliferation were investigated by CCK-8 method,and Hep G2 group( group C) and LX-2 and Hep G2 were co-culture group( group D)with common cell supernatants were set up. The influence of different concentrations HBs Ag on Hep G2 cells of Col I andα-SMA secretion were observed in all groups by ELISA method. Results With the increasing of the concentrations of HBs Ag,the proliferation of Hep G2 cells was significantly prompted in a concentrate-dependent manner in all groups.When concentration was 5 ng / ml,the percentages of Hep G2 cell proliferation were significantly higher than other groups,the difference was statistically significant( P〈0. 05). Compared with group C,the Col I and ɑ-SMA expression levels were increased in group A,there was statistically significant difference( t = 3. 419,P = 0. 005; t = 4. 388,P =0. 001). Compared with group D,the Col I and ɑ-SMA levels in group B were increased,there were statistically significant differences( t = 3. 048,P = 0. 009; t = 3. 005,P = 0. 009) after stimulation 48 hours by 5 ng / ml concentration HBs Ag. Conclusion HBs Ag can contribute to the proliferation of human liver cancer cells Hep G2. Human hepatocellular carcinoma line Hep G2 has characteristic of fibroblast cells with Col I and ɑ-SMA. HBs Ag may play a key role in promoting the progress of liver fibrosis,cirrhosis and liver cancer by stimulating Hep G2 secretion of Col I and α-SMA by stimulating Hep G2 and hepatic stellate cells LX-2.
