摘要
研究了磷对悬浮培养的西红花细胞生长、色素合成和主要营养物质吸收利用的影响。结果表明:不同初始磷浓度(低磷0.272 mmol/L、中磷0.544 mmol/L、高磷0.816 mmol/L)下西红花细胞均经历延滞期(0~3 d)、快速生长期(低、中磷3~15 d,高磷3~12 d)、稳定期(低、中磷15~21 d,高磷12~18 d)和衰亡期(低、中磷21~39 d,高磷18~39 d)这4个阶段,生长曲线呈S形。培养过程中比生长率呈下降趋势,磷对比生长率有一定影响。培养前9 d高磷下比生长率最高,中磷次之,9 d后高磷下的比生长率快速下降,中磷最高。进入快速生长期色素开始合成,到衰亡期色素仍继续合成,并且色素含量达到最高,随后色素开始降解,含量下降,色素合成期比细胞生长期长。因此,西红花细胞培养过程中色素合成与细胞生长不完全同步,属于部分耦联型。培养过程中色素比合成率呈上升趋势,磷对色素比合成率有一定影响。中磷浓度下色素比合成率最高。在快速生长期前期(6 d)质量分数95%蔗糖被降解为葡萄糖和果糖而被细胞吸收利用,培养18 d时质量分数85%葡萄糖被吸收利用,21 d时质量分数85%果糖被细胞吸收利用。培养3 d时胞外摩尔分数95%NH+4被细胞迅速吸收利用,而NO-3吸收利用较为缓慢,21 d时摩尔分数80%NO-3被吸收利用。提高初始磷浓度,果糖和NO-3的吸收利用速率亦随之提高,同时发现提高NO-3的吸收利用速率是提高色素合成能力的关键,高磷条件下细胞对NO-3的吸收利用速率显著提高。稳定期后期80%以上的营养物质被吸收利用,碳、氮源的不足影响了细胞生长,但对色素合成影响不大。
The effect of phosphate on cell growth,pigment biosynthesis and nutrients absorption and utilization in cell suspension culture of Crocus sativus L. was investigated. The results showed that at different initial phosphate concentrations( low phosphate 0. 272 mmol / L,middle phosphate 0. 544 mmol / L,high phosphate 0. 816 mmol / L) the saffron cells all went through four phases: lag phase( 0-3 d),rapid growth phase( low and middle phosphate 3-15 d,high phosphate 3-12 d),stationary phase( low and middle phosphate 15-21 d,high phosphate 12-18 d) and decline phase( low and middle phosphate 21-39 d,high phosphate 18-39 d). The growth curves were all S shapes. During the culture,specific growth rate turned to decrease,and phosphate had a certain effect on specific growth rate. In the first 9 days,specific growth rate was highest at high phosphate concentration,followed by middle phosphate. Then specific growth rate decreased rapidly at high phosphate concentration,and middlephosphate was the highest. Pigment biosynthesis started at rapid growth phase. At decline phase,pigment biosynthesis still continued,and which the pigment content reached the highest. Then pigment started to degraded and the content decreased. Pigment biosynthesis phase was longer than cell growth phase.Therefore,during the culture,pigment biosynthesis and cell growth were not fully synchronized. They were mixed-growth associated. During the culture,specific biosynthesis rate increased,and phosphate had a certain effect on specific biosynthesis rate. Specific biosynthesis rate was highest at middle phosphate concentration. During the culture,at the early of rapid growth phase( 6 d) mass fraction of sucrose of95% was degraded to glucose and fructose. Then they were absorbed. At 18 th day mass fraction of glucose of 85% was absorbed and at 21 th day mass fraction of fructose of 85% was absorbed. Mole fraction of NH+4of 95% was absorbed rapidly in the first 3 days.
