摘要
采用低聚木糖生产废渣替代麸皮固态发酵制备β-甘露聚糖酶,研究了碳源种类和组成、诱导物、氮源种类和组成对黑曲霉固态发酵制备β-甘露聚糖酶的影响。结果表明:低聚木糖废渣可部分替代麸皮固态发酵制备β-甘露聚糖酶。以低聚木糖废渣和麸皮质量比4∶6为复合碳源,2%的硫酸铵为氮源,黑曲霉在添加3%魔芋粉的培养基中于30℃条件下固态发酵5 d,制备的β-甘露聚糖酶、β-甘露糖苷酶活力分别为341.8、7.02μmol/(min·g),可溶性蛋白含量为8.40 mg/g。
The residues from xylooligosaccharides production was applied forβ-mannanase preparation instead of wheat bran, and effects of different kinds and ratio of carbon sources, inducer, nitrogen sources and loading on solid state fermentation forβ-mannanase preparation by Aspergillus niger were investigated in this work. Residues of xylooligosaccharides production could be instead of wheat bran partly forβ-mannanase preparation. Solid state fermentation of residues of xylooligosaccharides production and bran by A. niger for five days, which the mass ratio of residues of xylooligosaccharides production to wheat bran was 4∶6, 2% of ammonia sulfate and 3% of konjac flour loading in the medium, theβ-mannanase activity andβ-mannosidase activity were 341.8 and 7.02μmol/( min·g) , respectively, and soluble protein content was 8.40 mg/g.
