酶法协同超声提取紫苏叶总黄酮工艺条件优化 被引量:9

摘要 目的:采用酶法协同超声对紫苏叶总黄酮进行有效提取。方法:在单因素考察的基础上,以酶解p H、酶解温度、酶质量浓度、固液比、乙醇浓度、超声提取时间及超声提取温度为考察因素,以总黄酮提取率为指标,采用正交试验优选紫苏叶中总黄酮的提取工艺。结果:优化得到的工艺参数为:酶解p H 5.0,酶解温度40℃,酶质量浓度0.25 mg·m L-1,固液比1∶20,乙醇浓度70%,超声提取时间30 min,超声提取温度60℃。结论:酶法协同超声应用于紫苏叶总黄酮的提取,优选的工艺合理、可行。 Objective :To study a novel extraction process of flavonoids in Folium Perillae by cellulose enzyme com- bined with ultrasound. Methods: Based on the single factors research, the extraction technology was optimized by orthogo- nal design with the extraction rate of flavonoids as index and using pH value, enzymatic temperature, enzyme mass con- centration ,ratio to solid, the concentration of ethanol, ultrasonic time and ultrasonic temperature as factors. Results: The optimal extraction technology was as follows : pH 5.0, enzymatic temperature 40 ℃, enzyme mass concentration 0.25 mg · mL - 1, ratio to solid 1 : 201 the concentration of ethanol 70%, ultrasonic time 30 min, ultrasonic temperature 60 ℃. Conclusion: The flavonoids of Folium Perillae are extracted by cellulose enzyme combined with ultrasound and the opti- mized process is reasonable and feasible.
