ω-3鱼油脂肪乳对辐射损伤小鼠造血系统保护作用的研究 被引量:1

摘要 目的研究ω-3鱼油脂肪乳对电离辐射引起小鼠骨髓损伤的影响,旨在阐明ω-3鱼油脂肪乳对辐射性造血损伤的保护作用。方法采用直线加速器一次性6.0 Gy全身照射ICR小鼠,建立辐射损伤小鼠模型。取各组小鼠外周血用于白细胞(WBC)和血小板(PLT)计数,取骨髓进行瑞氏-吉姆萨染色观察骨髓象改变,流式细胞术检测骨髓细胞凋亡百分率,免疫细胞化学检测骨髓细胞凋亡抑制因子BCL-XL的表达。结果与空白对照组(BC组)相比,生理盐水组(NS组)小鼠外周血WBC和PLT计数均明显减少(P<0.05),骨髓增生降低,早期及晚期凋亡率显著增加(P<0.05),凋亡抑制因子BCL-XL表达降低;与生理盐水组相比,ω-3鱼油脂肪乳(ω-3组)可显著提高小鼠外周血WBC和PLT计数(P<0.05),促进骨髓增生,抑制骨髓细胞凋亡率(P<0.05),增加凋亡抑制因子BCL-XL的表达。结论ω-3鱼油脂肪乳可以促进骨髓造血系统恢复,对辐射导致的造血系统损伤具有良好的保护作用。 Objective To observe the effect of Omega-3 fish oil fat emulsion on mouse bone marrowinjury induced by ionizing radiation and to elucidate the protective effects of Omega-3 fish oil fat emulsion on hematopoietic injury caused by radiation. Methods The animal model of radiation injury was established by applying one dose of6. 0 Gy whole-body ionizing radiation by linear accelerator on ICR mice. The peripheral blood was collected to count white blood cell( WBC) and platelet( PLT). M arrowwas collected to observe the changes of bone marrowby WrightGiemsa staining. The apoptosis of bone marrowcell was detected by flowcytometry. The expression of BCL-XL in bone marrowcells was detected by immunocytochemical method. Results Compared with that in the blank control group( BC group),WBC and PLT count in peripheral blood of the normal saline group( NS group) significantly reduced( P〈0. 05),bone marrowhyperplasia reduced,early and late apoptosis rate increased significantly( P〈0. 05),BCL-XL decreased. Compared with that in NS group,omega-3 fish oil fat emulsion( omega-3 group) can significantly improve the peripheral blood WBC and PLT count( P〈0. 05),promote the proliferation of bone marrow,significantly inhibit the apoptosis rate of bone marrowcells( P〈0. 05),increase the expression of BCL-XL. Conclusion Omega-3fish oil fat emulsion can promote the recovery of the injured bone marrowhematopoietic system,exert a good protective effect on the radiation injury of hematopoietic system.
