有氧运动预防大鼠2型糖尿病过程中对机体铁代谢的影响 被引量:7

摘要 目的:探讨有氧运动预防高糖高脂饮食诱导大鼠2型糖尿病过程中,通过缓解机体铁超载,减轻氧化应激,改善胰岛素抵抗的机制。方法:24只12月龄SD大鼠,随机分为对照组(C组)、高糖高脂膳食组(H组)、高糖高脂膳食结合有氧运动组(HE组),每组8只。C组进行基础饲料喂养,其余两组使用高糖高脂饲料。HE组在2周适应运动后,进行15 m/min、45 min/天、坡度5度、6天/周、3周有氧运动干预,其余两组安静喂养。5周后,用酶联免疫吸附实验(ELISA)检测血清铁蛋白、血清胰岛素;生化试剂盒检测血清铁、血清总铁结合力、肝组织铁、血糖、肝超氧化物歧化酶(SOD)、肝丙二醛(MDA)、肝过氧化氢酶(CAT);肝组织切片用苏木精-伊红(HE)染色。结果:(1)HE组血清铁蛋白、血清胰岛素、血糖、胰岛素抵抗指数显著高于C组,显著低于H组(P<0.05,P<0.01)。HE组肝SOD、肝CAT、胰岛素敏感指数显著高于H组(P<0.05,P<0.01),H组显著低于C组(P<0.05,P<0.01);(2)HE组肝MDA、肝组织铁、血清铁、转铁蛋白饱和度显著低于H组(P<0.01)。(3)组织学观察表明,HE组较H组肝细胞脂肪滴数量少,无小灶状炎症细胞浸润。结论:(1)长期高糖高脂饮食诱导大鼠体重增加,加重铁过载和氧化应激,出现胰岛素抵抗。(2)有氧运动能够降低机体铁储存,增加抗氧化应激酶的活性,缓解氧化应激与胰岛素抵抗,减轻高脂膳食大鼠肝组织脂肪样变。 Objective To study the preventive effect of aerobic exercise on the type 2 diabetes mellitus.Methods 24 twelve-month old male SD rats were randomly divided into the standard diet control group(C,n=8),high-sugar and high-lipid fodder group(H,n =8),and high-sugar and high-lipid fodder plus exercise group(HE,n=8). The rats in group HE ran on a treadmill at the speed of 15m/min,45min/d,5-6d/w,for 3weeks,while in the other two groups were sedentary. The contents of serum ferritin and insulin were examined by ELISA kits 5 weeks after the experiment. Serum iron,total iron binding capacity,liver tissue iron,blood glucose,liver SOD,liver MDA,and liver CAT were examined by reagent kits. Liver tissue paraffin sections were stained with hematoxylin and eosin. Results(1)The amount of serum ferritin,serum insulin,blood glucose,and HOMA-IR in group HE were obviously higher than those in group C(P 〈0.05,P 〈0.01),while obviously lower than those in group H(P〈0.05,P〈0.01).(2)The amount of liver SOD,liver CAT,and HOMAISI in group HE were obviously higher than those in group H(P 〈0.05,P 〈0.01),in group H were obviously lower than those in group C(P 〈0.05,P 〈0.01). The amount of liver MDA,liver tissue iron,serum iron and transferrin saturation in group HE were obviously lower than that in group H(P〈0.01).(3)The number of fat drops in group HE was less than group H,and no inflammation appeared in group HE. Conclusion Aerobic exercise could reduce the body iron storage,elevate the activity of enzymatic antioxidants,release oxidative stress and insulin resistance,and thus alleviate the lipoid degeneration in liver of rats fed with high-sugar and high-lipid fodder.
