摘要
upp基因可用来作为反向筛选标记基因。利用p ORI温度敏感型双质粒系统敲除干酪乳杆菌Lactobacillus casei 393的upp基因,构建得到重组菌Lactobacillus casei 393-△upp。重组菌可作为反向筛选标记基因upp使用的宿主菌。
Upp gene exists widely in prokaryotes and some primary eukaryotes cells. UPRT ( Uracil Phosphoribosyl transferase ), eneoded by upp gene, can convert 5-fluorouracil to 5-fluoro-dUMP, which inhibits thymidylate synthase, causing cell death. So the upp gene can be used as a eounterse- lectable marker. In this study, we construst Lactobacillus casei 393-△upp mutant by pORI tempera- ture-sensitive dual plasmid system and the upp counterselectable marker can be used in this upp de- leted strain.
