摘要
以‘早酥’梨(Pyrus bretschneideri Rehd.)及其红色芽变‘红早酥’梨为试材,采用同源克隆方法,分别从这2种材料的果皮中,得到花青苷生物合成途径中3个关键结构基因PbCHS、PbDFR、PbUFGT和2个调控基因PbbHLH、PbMYB10的cDNA编码区序列。生物进化树分析表明,这5个花青苷合成相关基因序列与其他植物均具有较高的同源性。通过荧光定量PCR,研究花青苷合成基因在不同幼嫩组织器官中的表达情况。结果表明,大部分基因在‘红早酥’梨的幼叶、叶柄、花瓣、幼果、花梗中的表达量均高于‘早酥’梨,其中PbUFGT和PbMYB10在二者中的表达差异尤为显著。
In this study,cDNA coding region of three structural genes,PbCHS、PbDFR、PbUFGTand two transcription factors PbbHLH and PbMYB10 were isolated from peels of‘Zaosu'pear(Pyrus bretschneideri Rehd.)and it's red mutant(‘Hongzaosu'pear)using homologous cloning.Sequence alignment and phylogenetic analysis revealed that these genes shared high similarity with genes from other plants.Research gene expression in different young tissues by real time PCR,the result indicated that the majority of genes in‘Hongzaosu'pear showed higher transcript abundance than‘Zaosu'pear in young leaves,petioles,petals,young fruit,and stalks,which differentially expressed in both PbUFGTand PbMYB10 particularly significant.
