黑山羊醋酸钙不动杆菌的分离鉴定及生物学特性研究 被引量:2

摘要 为确诊南宁某养殖户黑山羊死亡的病因,取病死黑山羊的肝、脾和心血进行了细菌分离培养。通过形态学鉴定、生化试验、16S r DNA测序分析、小鼠攻毒试验、药敏试验和耐药基因检测等方法,对分离到的疑似致病菌进行鉴定及药敏特性观察。结果显示,分离菌为醋酸钙不动杆菌,可引起80%的小鼠死亡(4/5),表明该分离菌株对小鼠有较强的致病性。药敏特性发现该菌对青霉素类、头孢菌素类、呋喃类等4种抗生素耐药,对磺胺类、氯霉素类及多粘菌素类等5种抗生素呈中敏;对喹诺酮类、氨基糖苷类及大环内酯类等9种抗生素敏感。检出磺胺类、四环素类及氯霉素类等的14个耐药基因。结果表明,分离的醋酸钙不动杆菌毒力较强,有多重耐药性,对养殖业有一定的危害,要引起重视。 In this study, bacteria were isolated and cultured from the liver, spleen, and intracardiac blood of a dead black goat in order to identify the pathogen which had caused black goat death on a farm in Nanning. Then the suspected pathogens were ana- lyzed, tested and identified through morphological identification, biochemical tests, and 16S rDNA sequencing analysis, after which they were used to challenge mice to detect drug sensitivity and resistance gene. The results showed that the isolates were Acineto- bacter calcoacetlcus, with strong pathogenicity to mice, bringing forth 80% mortality rate (4/5). Detection of drug sensitivity dis- played that the strain had resistance to 4 types of antibiotics, such as penicillins, cephalosporins and furans, moderate resistance to 5 types of antibiotics, such as sulfonamides, chloramphenicol and polymyxins, and no resistance to 9 types of antibiotics, such as quinolones, aminoglycosides and macrolides. 14 genes were found to be resistant to Sulfonamides, tetracycline, chloramphenicol, etc.The results of this study indicated that the isolated Acinetobacter calcoacetlcus had strong pathogenicity. Being resistant to mul- tiple antibiotics, it can do harms to animal husbandry and aquaculture, and therefore should be paid due attention.
