摘要
目的:观察满山红总黄酮(Rhododendron dauricum L.total flavonoids,RTF)对肠平滑肌收缩功能的影响并探讨其作用机制。方法:采用炭末推进法,观察RTF对正常小鼠小肠蠕动的影响;采用离体小肠灌流技术,观察RTF对乙酰胆碱(Ach)、组胺和氯化钡所致肠平滑肌收缩的影响。结果:RTF(180、90、45 mg·kg^-1)可以剂量依赖性的降低炭末推进率(P〈0.01);RTF(0.5-4 g·L^-1)对乙酰胆碱(Ach)、组胺和氯化钡所致肠平滑肌收缩也具有剂量依赖性的抑制作用(P〈0.01)。在无钙灌流液中,RTF(2 g·L^-1)孵育10 min后可使CaCl2的量效曲线向下移位且可使Ach在无钙灌流液中诱导的肠管最大收缩幅度明显降低(P〈0.01)。结论:RTF在体内外能剂量依赖性的抑制小肠平滑肌的收缩,其机制与阻断M3受体和H1受体、降低细胞内游离钙离子浓度有关。
Objective : To observe effect of the Rhododendron dauricum L. total flavonoids ( RTF ) on the contraction of small intestine smooth muscle and explore the mechanism. Method: Carbo medicinalis facilitation method was used to observe effect of RTF on creeping of normal mice small intestine ; the isothermal perfusion of small intestine in vitro was used to observe effect of RTF on the contraction induced by Ach, histamine and BaC12. Result: RTF ( 180,90,45 mg· kg^-1 )decreased rate of earbo medicinalis facilitation with a dose-dependent manner ( P〈0.01 ); RTF ( 0.5-4 g ·L^-1 ) also inhibited the contraction of small intestine smooth muscle induced by Ach, histamine and BaCl2 with a dose-dependent manner ( P〈0.01 ). In Ca^2+ free K-H solution pre-depolarized with KC1, both Ca^2+ and Ach -induced contraction were dramatically attenuated after pretreated with RTF ( 2 g ·L^-1 )for 10 rain in krebs solution without Ca^2+ ( P〈0.01 ). Conclusion : RTF could inhibit the contraction of small intestine smooth muscle with a dosedependent manner in vivo and in vitro, and the mechanism may be related to antagonize the M3 and H1 receptor and reduce of mtracellular Ca concentration.
