摘要
目的本次研究通过建立大鼠急性胰腺炎模型,测定血清中IL-6、内毒素、TNF-α含量的变化以及HE染色观察胰腺组织的病理改变情况,探讨富氢液对急性胰腺炎的作用机制以及治疗效果。方法将45只大鼠随机分为3组,分别为假手术组(A组)、生理盐水组(B组)、富氢液组(C组),每组15只,禁食不禁水处理24 h后开始建立急性胰腺炎大鼠模型。富氢液组和生理盐水组大鼠胰腺发生水肿充血、局部呈现暗红色、并出现散在出血点时为建模成功,分别给予相同量的生理盐水和富氢液,假手术组仅轻微翻动胰腺几次,每组于第1、3、7天3个时间点分别取5只大鼠穿刺门静脉取全血2 ml,并切除胰腺组织,并于取材完毕后采用颈椎脱臼法处死相应大鼠,按照常规方法制作胰腺组织病理切片,HE染色后显微镜下观察大鼠胰腺组织病理变化,并采用鯊试剂偶氮显色法测定血清内毒素,采用酶联免疫法(ELISA)测定血清TNF-α、IL-6浓度,在整个实验过程中观察各组大鼠的一般反应情况,使用SPSS 13.0对实验数据进行分析。结果在各个时间点,B、C组大鼠内毒素、TNF-α、IL-6含量及病理学评分显著高于A组(P<0.05),且C组大鼠4种指标显著低于B组(P<0.05);A组大鼠反应比较灵敏,B组大鼠反应较为迟钝,而C组大鼠的反应与A组相对接近,与B组相比有明显的改善(P<0.05)。结论富氢液可显著降低急性胰腺炎大鼠体内的血清内毒素、TNF-α、IL-6含量,并减小胰腺组织的损伤,其机制可能与抑制脂质过氧化反应和炎症反应有关。
Objective This study through the established model of acute pancreatitis in rats, measuring the changes of IL- 6 in rat serum,endotoxin,TNF-α content and HE staining of pancreatic tissue were observed, and to explore the mecha- nism of hydrogen rich liquid on acutepancreatitis and the therapeutic effect. Methods 45 rats were randomly divided into 3 groups, respectively, the sham operation group( A group), normal saline group( group B) , hydrogen rich saline group( C group), 15 rats in each group, fasting. After 24 h of treatment, began to establish the model of acute pancreatitis in rats. Hydrogen rich water group and saline group rats pancreatic edema hyperemia,local dark red, and scattered bleeding points for modeling success, were given the same amount of saline and hydrogen rich water, sham operation group only slightly changes the pancreatic several times, each in first,3,7 days of 3 time point respectively,5 rats were portal vein puncture and take blood 2 ml, resection of pancreatic tissue, and by cervical dislocation were executed in after the corresponding rat pancreatic tissue pathological slices, made in accordance with the conventional method, pathological changes of pancreatic tissue of rats were observed by HE staining under the microscope, and plasma endotoxin was measured by colorimetry with shark azo reagent, using enzyme linked immunosorbent assay (ELISA) for determination of serum TNF-alpha, IL-6 concen- tration, observe the general response of rats in each group during the entire experiment, The experimental data were ana- lyzed using SPSS13.0. Results At each time point, the rats in group C, B,TNF-α lpha, IL-6 content of endotoxin and pa- thology scores were significantly higher than those in A group ( P 〈 0.05 ), and the rats in group C fourindex was lower than that in B group (P 〈 0.05 ) ; rats in group A reaction were more sensitive, the rats in B group were comparatively slow reaction,while the rats in group C and group A were relatively close, had the distinc
