大麦不同器官总RNA提取方法研究

摘要 [目的]探索出一套较好的大麦总RNA提取方法.[方法]以大麦的叶片、茎秆和子粒为试验材料,探索各器官总RNA提取方法.[结果]所得产物的检验结果表明,总RNA具有整齐且清晰的28S rRNA、18S rRNA条带,A260/A280均介于1.8 ~2.0.将提取的叶片总RNA反转录成cDNA,RT-PCR检测PCR产物在1 500bp处.[结论]用此方法提取的大麦各器官总RNA质量较好,纯度较高,完整性较好,可用于进一步的分子生物学研究. [ Objective ] The aim was to explore total RNA extraction methods for different organs of barley. [ Method ] Using barley leaves, stems and grains as the experimental materials , the total RNA extraction method of these organs were explored. [ Result] The detected results of these products showed that the total RNA had clear and neat stripes of 28S rRNA and 18S rRNA in the RNA electrephoresis. The ratios of A~/A2so were between 1.8 to 2.0. Leaves total RNA isolated by these methods were reverse transcribed and high quality cDNA were ob- tained. RT-PCR detected the product of PCR was in the 1 500 bp. [ Conclusion] These results showed that the quality of the extracted barley total RNA were good, high purity, good integrity, and were suitable for further research in molecular biology.
