摘要
首次提出了一种适用于小佛肚竹 (Bambusa ventricosa)和螺节竹 (Pleioblastus gramineus)等竹类植物总 RNA提取方法。用该方法提取叶片和笋尖的总 RNA具有正常的光谱吸收 ,OD2 60 /OD2 80 比值为 1.96~ 2 .0 ,OD2 60 / OD2 3 0 比值为 2 .96~ 4 .11;琼脂糖电泳后 ,2 8SRNA的亮度基本为18SRNA的亮度的 2倍 ;这说明提取的 RNA很完整 ,未降解 ,质量高。同时该方法操作简便、无DNA污染 ,且产率高 ;提取的 RNA可用于 RT- PCR、Northern杂交、c DNA文库的构建等。
A RNA extraction method applicable to Bambusa ventricosa and Pleioblastu gramineus leaves and tips of bamboo shoots was developed for the first time. The results showed that the total RNA from leaves and tips of bamboo shoots of B. ventricosa and P. gramineus had the normal spectrum of the ultraviolet absorption, the value of OD 260 /OD 280 varied between 1.96~2.0 and OD 260 /OD 230 varied between 2.96~4.11 by spectrophotometer, and the brightness of 28SRNA was approximately two times as light as that of 18SRNA in maps of electrophoresis. These indicated that the total RNA was very complete and did not degrade. With this method the pure RNA could be obtained simply,amply, economically without DNA contaminated, and be used for RT PCR, Northern blot and construction of cDNA library.
