布地奈德对哮喘大鼠气道重塑及气道平滑肌中Toll样受体4表达的影响 被引量:6

Effect of Budesonide on airway remodeling and TLR_4 of airway smooth muscle cell in rat models of asthma

摘要 目的:探讨布地奈德对哮喘大鼠模型气道重塑及平滑肌中Toll样受体4表达的影响。方法:将30只SPF级SD大鼠随机分为支气管哮喘组(A组)、健康对照组(B组)、布地奈德治疗组(C组)3组,每组10只。制备大鼠肺组织病理切片,HE染色后观察各组气道结构改变情况,并测定支气管壁厚度(WAt/Pi)、支气管壁平滑肌厚度(WAm/Pi)及支气管平滑肌细胞核数(N/Pi),利用Western-Blot方法检测气道平滑肌中TLR4的蛋白表达。结果:光镜下可见A组支气管上皮细胞脱落、管壁炎症细胞浸润、杯状细胞增生、基底膜和平滑肌明显增厚,而C组上述改变较A组明显减轻。与B组比较,A组支气管管壁厚度、平滑肌厚度及平滑肌细胞核数显著升高(P<0.01),与A组比较,C组支气管管壁厚度、平滑肌厚度及支气管平滑肌细胞核数显著降低(P<0.01)。A组的TLR4的蛋白水平明显高于B组及C组(P<0.01)。结论:吸入布地奈德可以防治气道重塑,可能通过抑制TLR4的表达实现。 Objective :To explore the influence of budesonide on the airway remodeling and TLR4 of airway smooth muscle cell in Rat models of asthma .Methods :Thirty SD rats were randomly divided into 3 groups :model group(group A) ,normal control group(group B) ,budesonide treatment group(group C) ,with 10 rats in each group . The airway inflammation and the alteration of airway structure were detected by HE staining ,The airway wall thick‐ness(WAt/ Pi) ,the bronchial smooth muscle thickness(WAm/Pi)and numbers of smooth muscle cell (N /Pi )were measured by using image analysis system .the expression of TLR4 protein in cells were detected by Western blot .Results :Bronchial epithelial desquamation ,inflammatory cell infiltration ,hyperplasia of goblet cells and smooth muscle cells were seen in group A under optical microscope ,however compared with group A ,the manifestations were signif‐icantly decreased in group C .Compared with group B ,WAt/ Pi ,WAm/Pi and N/Pi were expressed at higher levels in lung tissues of group A(P 〈 0 .01) .Compared with group A ,WAt/ Pi ,WAm /Pi and N /Pi were expressed at lower levels in lung tissues of group C(P〈 0 .01) .Compared with group B and C ,the expression of TLR4 at protein levels in group A was significantly highter .Conclusion :The airmay remodeling can be inhibited by budesonide proba‐bly through reducing the expression of TLR4 .
