人参缩心饮通过β2肾上腺素受体对心衰大鼠心肌细胞收缩功能的影响 被引量:4

Myocardial contractile function of Ginseng Drink for shrink heart by affecting the β2-AR cells in heart failure rats

摘要 目的:探讨人参缩心饮是否具有影响心衰大鼠心肌细胞收缩功能。方法:异丙肾上腺素制作大鼠心衰模型并分离培养心肌细胞。将心肌细胞分为:正常心肌细胞组和心衰心肌细胞组。正常心肌细胞组的培养分别为正常高糖培养基(DMEM)和含50nmol/Lβ2肾上腺素受体(β2 adrenaline receptor)拮抗剂(ICI118551)的DMEM培养基,心衰心肌细胞组的培养条件分别为:DMEM培养基,含有100nmol/L人参缩心饮的DMEM培养基,含有50nmol/L ICI118551和100nmol/L人参缩心饮的无血清的DMEM。分别测定心肌细胞收缩幅度和存活率,Western blotting测定心肌细胞β2AR蛋白表达。结果:与正常心肌细胞组相比,心衰心肌细胞组收缩幅度和存活率均显著降低(P<0.05),心衰心肌细胞组心肌细胞在使用β2AR拮抗剂后,收缩幅度和存活率进一步降低(P<0.05);线性回归分析显示β2AR蛋白表达与心肌收缩幅度呈正相关(R2=0.9656)。但使用人参缩心饮后,心衰心肌细胞收缩幅度和存活率均出现显著升高(P<0.05),β2AR蛋白表达增加;人参缩心饮干预后,心衰心肌收缩幅度与β2AR蛋白表达呈正相关(R2=0.9911)。结论:人参缩心饮通过作用于β2AR增强心肌细胞收缩率和存活率,更好地维持了心脏的正常功能。 Objective: This paper aims to investigate whether the Ginseng Drink have the contractile function effect on the myocardial cells of heart failure rats. Methods: Isoproterenol produced heart failure in rats and isolated and cultured cardiomyocytes. The myocardial cells were divided into: normal cardiac myocytes cell group and heart failure group. Normal cardiac myocytes cell groups were separately cultured in normal DMEM medium and containing 50nmol/L β2-AR antagonist (ICI118551) DMEM medium. The culture conditions of failure cardiomyocytes group separately were DMEM medium, containing 100nmol/L DMEM medium, containing 50nmol/L β2-AR antagonist (ICIl18551) and 100nmol/L Ginseng Drink DMEM. Myocardial contraction amplitude and survival were measured, β2A protein of cardiomyocytes were measured by Western blotting (WB). Results: Compared with normal cells, cell contraction amplitude and survival of heart failure were signifcantly lower (P〈0.05).The contraction amplitude and survival of failure myocardial cells were further reduced after using β2AR antagonists (P〈0.05). Linear regression analysis showed 13 2AR protein expression and cardiac contraction amplitude was positively correlated (R^2=0.9656). But after using Ginseng Drink, the contraction amplitude and survival rate of failure myocardial cells both significantly increased (P〈0.05), the protein expression of β2AR still increased. At the same time, linear regression analysis the showed that contraction amplitude and the protein expression of β2AR was positively correlated (R^2=0.9911).Conclusion: Ginseng Drink can enhanced myocardial cell contraction amplitude and survival by acting on adrenaline β2-AR to maintain the normal function of the heart.
