摘要
目的探讨不同浓度丝裂霉素C外用对兔耳增生性瘢痕的抑制效果。方法 18只日本大耳白兔,耳腹侧制作兔耳增生性瘢痕模型,创口按随机数字表法分生理盐水组(A组)、0.1 mg/ml丝裂霉素C组(B组)、0.05 mg/ml丝裂霉素C组(C组)、1 mg/ml曲安奈德组(D组),用相应溶液外用兔耳新鲜创面,局部保留,彩色多普勒观察创面愈合后增生性瘢痕厚度,并切除瘢痕组织,病理切片HE染色,显微镜(×200倍)下计数单个视野下的成纤维细胞数。结果 A组瘢痕厚度及成纤维细胞数均高于B、C、D组,C组瘢痕厚度及成纤维细胞数高于B组,差异均有统计学意义(P<0.05)。B组和D组比较差异无统计学意义(P>0.05)。B组和C组切口未见明显坏死和延迟愈合。结论外用丝裂霉素C能够抑制成纤维细胞生长,抑制兔耳瘢痕增生,并且不会造成局部组织坏死,具有较高的安全性。
Objective To observe the inhibitory effect of different concentrations of mitomycin C on rabbit ear hypertrophic scar.Methods Rabbit ear hypertrophic scars model was established in 18 Japanese white rabbits .The animals were randomly divided into four groups:normal saline group(A),0.1 mg/ml mitomycin C group(group B),0.05 mg/ml mitomycin C(group C) and 1 mg/ml triamcinolone group(group D).The fresh wounds of the animals were externally given and kept with the corresponding treated solutions in each group.Then,the thickness of hypertrophic scar after wound healing was counted with a color Doppler .Further,the scar tissues were removed for HE staining,and fibroblast cells under a single vision were counted with a microscopy (×200 times).Results The scar thickness and fibroblast cells in the group A were significantly greater than that in the groups B , C or D ( P <0.05 ) .The scar thickness and fibroblast cells in the group C were also significantly greater than that in the group B ( P <0.05 ) .There was no signifi-cant difference between the group B and group D .No incision necrosis and delayed healing was found in the group B and C .Conclusion External application of mitomycin C can inhibit the growth of fibroblast cells and scar hyperplasia in rabbit ears and does not induce the local tissue necrosis .Thus,it has a relatively high safety .
