4-羟苯基维胺联合超声对兔耳增生性瘢痕组织细胞增殖抑制作用的研究 被引量:3

摘要 目的 :采用兔耳增生性瘢痕模式探讨4-羟苯基维胺(HPR)联合应用超声波对增生性瘢痕组织细胞的增殖抑制作用,探索防治增生性瘢痕的新方法。方法:建立兔耳增生性瘢痕模型,分别设立对照组、4-HPR组、超声组、4-HPR+超声组,增生性瘢痕形成后分别进行治疗。4-HPR浓度分为2μmol/L、20μmol/L,超声强度分为超声有效声强为0.5 W/cm2,脉冲持续2ms(P2档);超声有效声强为0.75 W/cm2,脉冲持续3 ms(P3档);超声有效声强为1.25 W/cm2,脉冲持续5 ms(P5档),分别照射每个部位持续1、3和5 min,每7 d治疗1次,共进行4次。治疗结束1周后,经手术采集瘢痕组织,添加Brd U进行培养3 h,用10%甲醛固定,石蜡切片,用Brd U免疫组化的方式,检测真皮层成纤维细胞的增殖效应。结果:与对照组相比,4-HPR和超声照射对增生性瘢痕组织真皮层成纤维细胞增殖活性的抑制具有统计学意义(P<0.05),2μmol/L 4-HPR的低剂量联合应用超声波时差异也有统计学意义(P<0.01)。结论:4-HPR以及超声联合治疗能抑制增殖性瘢痕组织真皮层成纤维细胞的增殖。 Objective: To investigate the inhibitory effects of 4-hydroxy phenyl retinamide(4-HPR) combined with ultrasonic treatment on the proliferation of hypertrophic scar in rabbit ears, and to explore new preventive and therapeutic approaches for hypertrophic scars. Methods: Hypertrophic scar was established on rabbit ears. The rabbits with hypertrophic ears were divided into 4 groups. Animals in 4-HPR groups were treated with 2 μmol/L or 20 μmol/L 4-HPR. Animals in ultrasonic treatment group were treated with ultrasound at the intensities of 0.5 W/cm2(P2, 2 min), 0.75 W/cm2 (P3, 3 min), and 1.25 W/cm2 (P5, 5 min). Additional group of animal were treated with both 4-HPR and ultrasound while untreated animal served as controls. Treatments were performed once a week for 4 weeks. One week after the last treatment, the scar biopsies were taken and incubated with BrdU for 3 hours. Following fixation with 10% formalin, dermal fibroblast proliferation was evaluated using BrdU immunohistochemical technique in paraffin sections. Results: Compared with the control group, both 4-HPR and ultrasonic treatment markedly inhibited dermal fibroblast proliferation in hypertrophic scar (P〈0.05). Moreover, low dose of 4-HPR (2 μmol/L) combined ultrasonic treatment also significantly inhibited fibroblast proliferation (P〈0.01). Conclusion: These results show that 4-HPR combined ultrasonic treatment can inhibit fibroblast proliferation in the dermis of scar tissues.
