摘要
目的探讨氯化镉对人肾小管上皮细胞(HK-2细胞)形态及生存率的影响。方法用DMEM培养液传代培养HK-2细胞,生长至70%~80%融合时随机分成6个组,A、B、C、D、E组分别加入5μmol/L、10μmol/L、20μmol/L、50μmol/L、100μmol/L氯化镉终浓度,每组2个平行细胞瓶,继续培养24 h,终止培养。F组不加氯化镉,作为对照。培养24 h后观察细胞形态的变化,并采用MTT试验检测各组光密度值(OD值),计算细胞存活率。结果对照组、A组、B组的HK-2细胞形态均无明显变化;C组HK-2细胞开始发生形态学变化,细胞活力下降。C组、D组、E组HK-2细胞存活率低于对照组(P〈0.01),A组、B组HK-2细胞存活率与对照组比较差异无统计学意义(P〉0.05)。结论 20μmol/L浓度以上的氯化镉对HK-2细胞造成损伤,致细胞形态破坏,细胞存活率降低。
Objective To explore the effect of cadmium chloride on the morphology of human renal tubular epithelial cell( HK-2 cell) and survival rate. Methods The HK-2 cells cultured with DMEM medium were randomly divided into six groups when they reached 70% to 80% fusion. Groups A, B, C, D, E were given 5,10,20,50,100μmol/L of cadmium chlorine, respectively, with two parallel cell bottles in each group,and the culture lasted for 24 hours. Group F( control group) wasn't given cadmium chlorine. The changes in cellular morphology were observed after 24 hours of incubation. Methyl thiazol tetrazolium(MTF) assay was used to detect the optical density(OD) value in each group, and the survival rates of the cells were calculated. Results The morphology of HK-2 cells showed no significant difference among control group, group A and group B. The HK-2 cells appeared morphological changes in group C,with the decreased cell viability. The survival rates of HK-2 cells in groups C ,D,E were significantly lower than that in the control group(P 〈0.01 ). The survival rates of HK-2 cells in groups A, B showed no statistically difference compared with control group ( P 〉 0.05 ). Conclusion Cadmium chloride with the concentration of 20 μmol/L or more might lead to HK-2 cell injury,damages to cellular morphology and decreased cell viability.
