摘要
为研究木薯野生近缘种与栽培种间光合特性的差异,选用野生近缘种W14与栽培种华南8号(SC8)为材料,测定其叶绿素含量,同时采用便携式LI-6400光合作用测定仪,测定2种木薯叶片的净光合速率(Pn)、气孔导度(Gs)、胞间CO2浓度(Ci)、蒸腾速率(Tr)等光合参数,采用Western Blot对光合作用相关蛋白质核酮糖-1,5-二磷酸羧化酶/加氧酶(Rubisco)、放氧复合体(OEC)及D1的表达水平进行分析,并探讨其叶片光合特性的差异。结果显示:SC8叶片中叶绿素a及总叶绿素含量、Pn、Gs均显著高于W14,而叶绿素b含量、Ci及Tr在两者间并无显著差异,Rubisco、OEC及D1在SC8中的表达水平均高于W14。叶绿素含量、Pn及参与光合作用相关蛋白质表达水平的分析结果表明,栽培种SC8具有较好的光合特性。
In order to study the photosynthetic characteristics difference between cassava wild species and cultivated species, wild relatives of W14 and cultivated species of South China 8 (SC8) were as the materials. Chlorophyll content were measured, and net photosynthesis photosynthetic rate (Pn), stomatal conductance (Gs), intercellular CO2 concentration (Ci), transpiration rate (Tr) were also measured using a portable analyzer LI-6400, expression level of ribulose -1,5- two phosphate carboxylase/oxygenase (Rubisco), the oxygen evolving complex (OEC) and D1 which related with photosynthesis were analyzed using Western Blot, the differences of photosynthetic characteristics between W14 and SC8 were investigated. The results showed that: chlorophyll a, total chlorophyll, net photosynthetic rate (Pn) and stomatal conductance (Gs) in SC8 were significantly increased, while chlorophyll b, intercellular CO2 concentration (Ci) and transpiration rate (Tr) were no significant difference compared with W14, the expression level of Rubisco, OEC and D1 in SC8 were higher than those in W14. The results of chlorophyll content, net photosynthetic rate and expression level of proteins related to photosynthesis showed SC8 had better photosynthetic characteristics compared with wild species.
