摘要 以德国鸢尾(Iris germanicaL.)品种“金娃娃”当年新萌发幼芽的茎尖为外植体,进行了组织培养和快速繁殖技术的研究。结果表明:使用0.1%HgCl:对外植体消毒7min的效果最佳;初代培养、继代培养、生根培养最适合的培养基分别是MS+6-BA1.5mg/L+IAA0.2mg/L、MS+6-BA2.0mg/L+IAA0.5mg/L、1/2MS+6-BA0.2mg/L+IAA1.5mg/L;采用含4-5个芽的芽丛进行继代培养的增殖系数最高;试管苗移栽的最佳基质为珍珠岩+草炭+园土(1:1:1),移栽成活率达到91.6%。 The stem tips of newly-germinated tender buds of the German iris (Iris germanica L. ) variety "Gold boy" were taken as explants, and the tissue culture and rapid propagation techniques of iris were studied. The results showed that disinfecting the explants with O. 1% HgC12 for 7 min was the most effective. The optimal media for the primary culture, subculture and rooting culture of iris were MS +6-BA 1.5 mg/L+IAA 0.2 mg/L, MS+6-BA 2.0 mg/L+IAA 0.5 mg/L, and 1/2MS+6-BA 0.2 mg/L+IAA 1.5 rag/L, respectively. The subculture of bud cluster with 4 - 5 buds obtained the highest propagation coefficient. The optimum sub- strate for the transplanting of iris test-tube plantlets was perlite+ turf+ garden soil ( 1:1:1 ), and the survival rate after being trans- nlanted reached 91.6%.
