茶多酚对缺氧诱导的肺高血压大鼠肺动脉压和凋亡抑制蛋白表达的影响 被引量:3

摘要 目的探讨茶多酚对缺氧诱导的肺高血压大鼠肺动脉压和凋亡抑制蛋白Livin、X相关凋亡抑制蛋白(XIAP)表达的影响。方法将24只SD大鼠随机均分为正常对照组(C)、缺氧组(H)和茶多酚组(TP)。缺氧前TP组大鼠每日予茶多酚[200 mg/(kg·d)]灌胃,C组和H组大鼠接受等体积生理盐水灌胃,随后TP组和H组大鼠置于常压间断缺氧舱(8 h/d,每周6 d)4周。4周后比较各组的平均肺动脉压(mPAP)、右心室肥厚指数(RVHI)及肺小动脉管壁厚度,比较肺动脉平滑肌细胞(PASMCs)凋亡及肺组织Livin和XIAP表达水平。结果 H组大鼠的mPAP、RVHI、肺小动脉管壁厚度均比C组显著升高(P<0.01),且PASMCs凋亡明显减少(P<0.01),肺组织XIAP mRNA表达显著升高(P<0.01)。TP组mPAP及RVHI与H组相比均明显降低,肺小动脉管壁增厚程度减轻,而PASMCs凋亡增加(P<0.05),肺组织XIAP mRNA表达降低(P<0.05)。大鼠肺组织的Livin mRNA表达各组差异无统计学意义(P>0.05)。结论茶多酚可改善缺氧诱导的大鼠肺高血压,其机制可能与抑制肺组织XIAP表达水平,促进PASMCs凋亡有关。 Objective To investigate the effects of tea polyphenols(TP) on pulmonary artery pressure and the inhibition of apoptosis proteins in rats with hypoxia induced pulmonary hypertension.Methods Twenty four Sprague Dawley SD rats were randomly divided into three groups (n= 8 in each group):Normal control group(C),hypoxia group(H)and TP group.Before the hypoxia state, the rats in TP group were given TP 200 mg/(kg· d)each day.The rats in C group and H group were given normal saline.Then the rats in TP group and H group kept in atmospheric pressure with intermittent hypoxia for 8 hours a day,6 days weeks.After 4 weeks, mean pulmonary artery pressure mPAP,right ventricular hypertrophy index RVHI,wal thickness of smal pulmonary artery were compared.Apoptosis of pulmonary arterial smooth muscle cel s(PASMCs)and the mRNA expression of X linked inhibitor of apoptosis protein XIAP and Livin were also detected.Results Compared with C group,mPAP,RVHI and wal thickness of smal pulmonary artery were increased in H group (al P〈0.01),decrease in smooth muscle cel apoptosis (P〈0.01),and increase in XIAP mRNA expression(P〈0.01).There were decrease in mPAP,RVHI,wal thickness of smal pulmonary artery in TP group compare with H group,increase in smooth muscle cel apoptosis (P〈0.05)and reduction in XIAP mRNA expression (P〈0.05)in TP group.However,Livin mRNA expression was not significantly different in three groups(P〉0.05).Conclusion TP ameliorate hypoxia induced pulmonary hypertension in rats,its mechanism may be related to inhibit XIAP but not Livin expression,promoting the apoptosis of PASMCs.
